We screened pipe mutant with the treatment of EMS in Arabidopsis. Through observing the phenotypes of pipe mutant, we found that the seedling of pipe was dwarf and lost apical dominance. The siliques of pipe showed crook growth. Compared with wild-type, the shape of pipe leaves were obviously narrow and anomalistic curled. The PIPE gene was suggested to encode a protein phosphatase, our lab has found that PIPE is involved in the GA signaling transduction.Then the pipe mutants were treated with EMS secondly to establish a new mutant pool which containing the pipe mutant site. This mutant pool concludes 975 lines. Judging by the plant height, crook siliques, curled leaves and the shape of leave pavement cell, we screened 71 lines which rescued the pipe phenotype completely or partially and 10 lines which aggravated the pipe phenotype. Some of these lines were completely rescued the pipe phenotype and displayed other interesting phenotypes. For example,7151 and 7631 lines showed defects in flower organs. This remutagenesis pool provides abundant material and genetic evidences to further understand the PIPE function.We studied one line 3432 pipe from the mutant pool. The phenotype of 3432 pipe is similar to wild-type and rescued the pipe phenotype completely. The water lose rate of 3432 pipe was high and the drought tolerance of it was reduced. The stomatal movement of 3432 pipe was insensitive with the treatment of ABA, but the germination and growth of it were sensitive to ABA. The response level of exogenous PAC and GA3 in 3432 pipe is similar to pipe. The transcription levels of the key genes in GA biosynthetic pathway in 3432 pipe were also similar to pipe. Through the detection with the antibody of RGA and GAR, we found that the contents of RGA and GAI in 3432 pipe were also closed to that of pipe. The 3432 pipe mutant site was mapped to a 550 Kb genomic region by map-based cloning.
