This paper introduces the concept, characteristics, history, generation mechanismand application prospects of bio-photon radiation. The paper designed and developed abiological ultra-weak luminescence detection system. The system mainly consistsdarkroom, built-in excitation light source, light filter, shimmer acquisition module, thecontrol circuit and microcomputer. Excitation light source consists the singlehigh-power light-emitting diode (LED) and lenses, ensure that the sample subjected to auniform illumination. LED is powered by a precision numerical controlled constantcurrent source, to ensure the stability of the light source, while also easy to adjust thebrightness of the excitation light source. A filter is set between the sample and thedetector in order to change wavelength of light. And the luminescence spectrum of thesample can be measured indirect by measuring the delayed light emission curve of thesample, so as to avoid the spontaneous emission is too weak to be measured. Lightacquisition module using photomultiplier tube (PMT) as the core of a serial singlephoton counting system. IN order to protect the photomultiplier tube from beingdamaged by light of excitation light source, a shutter has installed before thephotocathode. When the excitation light source irradiate the sample, the shutter closes,protecting the photomultiplier tube; when the light turns off, the shutter opensimmediately, photomultiplier tubes began collecting samples delayed luminescencesignal. System uses microcontroller chip as the core controller, controls the outputcurrent, excitation light illumination time, opening and closing of the shutter and otherinput/output operations such as LCD displaying and keyboard scanning.The system hashigh precision, repeatability and can accurately collect biological super-weakluminescence signal, which has been proofed by measuring the gold heartChlorophytum leaves delayed luminescence signal.The paper also tested the basic features of ultra-weak luminescence spectra fornormal germinated mung bean seeds by using independently developed ultra-weakluminescence test system. It was found that the luminous intensity of germinated seedsis different in different wavelength ranges. This also shows as a more detailed analysismethod, spectroscopic analysis can be used to study the biological ultra-weakluminescence.
