Micro-fluidic chip has many advantages ,such as,less sample needed, low cost, rapid analysis, high integrability, chemical analysis. Therefore, it has a very wide range of applications in biological analysis, environmental monitoring, and clinical diagnosis. Meanwhile, the micro-fluidic chip technology using in blood analysis is one of the most routine clinical analysis, which is played a very important role in diagnosis and treatment of disease, which is also widely concerned. This thesis committed to research micro-fluidic chip production process, and then uses it in the Red Blood Cell (RBC) Deformability analysis research. This research includes three aspects:The first part, we repeated the experiments of PDMS molding process, to improve the existing molding process. And we also proposed a new fabrication technique of PDMS chips under the condition of equipment.The second part of the innovation has made a double-channel of PDMS molding method. This method can solve the micro-fluidic chip system layout and integration of functional modules to provide a good direction. While this method can not only be applied to micro-structure of the system integration chip design, but also be a rich manifold structure and function of chip design. At the same time this method is simple and has good standards and high repeatability.The third part, we used image analysis technique to measure flow velocity. It will simplify the micro-fluidic chip measurement system. And describe speed of response value directlyThe fourth part, we designed a molding to make sure the chips have the same size, which is called standardized production, and then improved the inlet to reducing the required amount of samples .which could be used in clinical testing. In this thesis we experiment many times with this improved micro-channel array RBC deformation detection system. The result shows that the improved detection system was repeated repeatability and reliability. The system provides information quickly and accurately, can be wildly used for the micro-channel measurement of RBC deformability in the promotion. Moreover, the achievements of PDMS molding production will direct a bright prospect in PDMS micro-chip.
