In recent years, much interest in the development of magnetic bead (MB) applicationsarises in microfluidic chips. MB is widely used in biomedical engineering for its highsurface area, coupling capacity and convenience of being manipulated. MB-basedbio-microsystem, which is designed for bimolecular manipulation, has become one of themost attractive research points inĪ¼TAS.In the research on MB microsystem based on MEMS technology, the design of a MBchip, manipulation of MB and applications of bio-reaction are the key process. This thesisstudies the rapid fabrication process of a novel high-gradient magnetic field microchip forMB separation. The chip is applied in analysis of DNA and proteins. Combined with MBtechnology, the total analysis from cell lysis to DNA amplification is achieved to realize ona single chip.The topics of this dissertation are presented below:1. MB technology and its applications in biomedical engineering are reviewed. Researchscheme is elicited on the basis of the investigation about achievements concerning MBtechnology and its microsystem.2. Based on the principle of MB capture, a high-gradient magnetic field device isintroduced for MB manipulation. COMSOL MYLTIPHHYSICS and MATLAB areused to simulate the velocity field and magnetic field, all of which provide efficientguidance for chip design and fabrication.3. Focus on the study of rapid fabrication process of the integrated micro magneticcolumn which is the functional structure of high-gradient magnetic field separation chip.4. Using multilayered PDMS patterning and MB techniques, an integrated microfluidicdevice with on-chip performance of cell lysis, DNA extraction and PCR amplificationis achieved to realize the total analysis from cell lysis to DNA amplification on a singlePolydimethylsiloxane (PDMS) chip.5. Combined with ELISA techniques, the MB chip is applied in AFP detection.This thesis studies simulation and analysis towards the magnetic field of a novelhigh-gradient magnetic field chip, and its rapid fabrication process, all of which willimprove the construction of MB microsystem. DNA analysis from thermal cell lysis, solidphase extraction of DNA and on-chip Polymerase chain reaction is realized on a singlechip, which shows an approach towards an integrated micro system capable ofcomprehensive genetic analysis on single chip.
