Magnetic Bead (MB) is widely used in biomedical engineering for its high surface area, coupling capacity and convenience of non-touch manipulation. Combined with microfluidic technologies, research on MB-based bio-microsystem has become one of the most attractive fields inμ-TAS, which can be widely used in cell separation, immunoassay and DNA extraction.This dissertation focuses on a novel MB bio-microsystem based on microfluidic technology. The topics of this dissertation are presented below:1. Introduction of MB technology and its applications in biomedical engineering. Eliciting research scheme based on the investigation of achievements concerning MB microsystem.2. Theory of the MB microsystem was studied. Modeling and Simulation of the magnetic field regenerated by the permalloy microelements was performed. The feasibility of magnetic capture has also been discussed.3. Process design of the MB microsystem. A fully transparent microsystem based on permalloy micro-electroplating and PDMS/Glass bonding was well constructed and a macro-to-micro interface process was developed for the system.4. On-chip experiments. On-chip operation of magnetic beads has been testified successfully and some experiments have been developed to realize the real-time, on-chip separation of some biological samples including proteins andλDNA products.The dissertation primarily provides a systematic approach towards the MB bio-microsystem based on microfluidic technologies.
