Cell Culture Chip system is a newly developed research field. It mainly consists of a cell culture chip, a thermal control apparatus, and a set of culture media perfusion apparatus. The micro-channels, micro-chambers on the chip are fabricated by using MEMS technology in the substrate of glass, silicon, polymer etc. The system has been applied for cell migration, cell differentiation, drug screening etc.In this dissertation, Cell Culture Chip system has been systemically studied, including chip designation, microfabrication, temperature controll and cell culture on chips.The topics of this dissertation are presented as below:1. Research and development on cell culture chips have been reviewed. Then new projects are carried out.2. Cell culture chips integrated with heaters are designed based on the underetch phenomenon in glass wet etching.3. Designation and construction of a peripheral thermal control system for integrated cell culture chips. The thermal controller has advantages of small volume, easy operation and real-time accurate temperature sensing and control.4.The effects of temperature adjustment, gas supply, culture media improvement and perfusion are studied. Porcine iliac artery endothelial cell line (PIEC) and 3T3 fibroblast cell line on-chip culture have been demonstrated.The dissertation shows that Cell Culture Chip system has some advantages such as low sample consumption, cell observation and record in vitro for a long time. But how to decrease the cost of Cell Culture Chip system and to enhance its stability need further works.
