In this paper, a theory about the magnetic beads adsorption in microfluidic chip was analysed, and by using of the simulation method the trajectories of magnetic beads under the permanent magnet adsorption was obtained, at last by combining with experiment, the movement trends of the magnetic beads under the influence of the external factors was verified.First, according to the stress analysis of the beads in the coupling of the magnetic field and the flow field, combining with the knowledge of theoretical mechanics, fluid mechanics, and electromagnetics, the magnetic force and the fluid drag force were obtained, and the motion equations of the magnetic beads were established, by using of simulation software COMSOL the numerical calculation of the magnetic field that generated by the permanent magnet and the fluid flow field were established, got the relevant parameters that affect the motion equations of the beads including magnetic field strength, magnetic field magnetic scalar potential, as well as the flow field velocity, and on this basis, the simulation trajectory of the magnetic beads in the coupling field was obtained. Combined with the equations of motion, these factors that affecting the movement of beads including of fluid flow rate, fluid viscosity, the radius of the bead, bead susceptibility, and permanent magnet residual magnetization of the permanent magnet and micro-flow channel spacing were analysed, and through change these parameters, the effect trends of the beads adsorption were obtained, and got the corresponding values of these parameters that affect the adsorption rate of the magnetic beads in the adsorption area.On the basis of theoretical studies, a microfluidic chip that the permanent magnetcan embed in it was produced, the permanent magnet can be extracted from the chip, and will not damage the chip, the purpose is to improve the utilization of the chip. Initially observational test of the flow rate that affect the adsorption rate of magnetic beads was carried out, the obtained image was processed by using of the MATLAB software, got the adsorption trend of the beads under different flow rates.And compared with the influence that the flow rate on magnetic beads adsorption rate in the simulation, and got the consistency effects that on the magnetic beads adsorption tendency. And compared with the beads adsorption trends in simulation,in order to verify consistency of the two pairs in beads adsorption trends.
