Object: To explore the association of BCAS3 single nucleotide polymorphisms(rs11653176and rs9905274) with gene expression in gout.Methods: 152 patients with acute gout, 67 patients with non-acute gout and 280 gout-free controls were recruited from the Affiliated Hospital of the Medical College, Qingdao University. Peripheral blood mononuclear cells BCAS3 m RNA and serum IL-1b were measured in different genotype carriers and different group. Genotyping of BCSA3rs11653176 and rs9905274 was performed by Taq Man? allele discrimination assays. An association analysis was carried out using the and A genotype–phenotype analysis χ2 test.Results: Our results showed that polymorphisms of rs11653176 and rs9905274 in BCAS3 were associated with gout in Chinese Han male population. BCAS3 m RNA were significantly increased in both acute gout patients and non-acute gout patients(P=0.06,0.004). BCAS3 m RNA in three genotype of rs11653176 were significantly different in both acute and non-acute gout patients(P=0.024,0.014);BCAS3 m RNA and IL-1βin three genotype of rs9905274 were significantly different in acute gout patients also(P=0.038,0.017).Conclusion: Our study suggested that the polymorphisms of BCAS3 may be relevant host susceptibility factors for the development of gout in Chinese Han male population. BCAS3 single nucleotide polymorphisms can affect the gene expression, and might participate in regulating immune and inflammation. However, further study should be done in a larger size sample to test and verify our result.
