| Objective: To investigate the genetic association between the single nucleotide polymorphisms(SNPs) of rs1800469, rs1800470, and rs4803455 in transforming growth factor beta 1(TGF-β1) gene and gout susceptibility in Chinese Han male population.Methods: A total of 507 Chinese Han male patients with gout and 526 matched healthy male controls were included in this study. SNPs were genotyped by using Taq Man SNP Genotyping Assays. Chi-square testing was adopted to compare differences in genotypic and allelic frequencies. Furthermore, the associations between different genotypes and the genetic susceptibility to primary gout were analyzed.Results:(1) The differences of the genotypes of CC, CT, TT in rs1800470 in patients(23.08%, 47.14% and 29.78%) and controls(30.04%, 45.82% and 24.14%) were significant(P=0.020). The differences of the allelic frequencies of C, T in patients(46.65%, 53.35%) and controls(52.95%, 47.05%) were also significant(P=0.004). Those carrying genotype TT had a higher risk for gout than those carrying genotype CC(OR=1.641,95%CI 1.181-2.286,P=0.009, after the adjustment of body mass index,waist-hip ratio, systolic blood pressure, fasting plasma glucose, triglycerides and creatinine).(2) In gout patients group, the differences of the genotypes of CC, CT, TT and the allelic frequencies of C, T in rs1800470 in tophi group and no tophi group were significant(P=0.022,P=0.006). Those carrying genotype TT had a higher risk for tophi than those carrying genotype CC(OR=2.661,95%CI 1.322-5.376,P=0.010, after the adjustment of serum uric acid and duration).(3) There was no difference in genotypes or allelic frequencies in rs1800469 and rs4803455 between patients and controls(P >0.05). There was no difference between tophi group and no tophi group, either(P>0.05).(4) For polymorphism rs1800470, durations and serum triglycerides levels showed significantly different distributions among the TT, CT, and CC genotype groups(P=0.045,P=0.001). But there was no difference when compared with other indexes(P>0.05).Conclusion: The polymorphisms of rs1800470 in transforming growth factor beta 1 gene are probably associated with the development of gout in Chinese Han males. Moreover,they may be closely related to the formation of tophi. However, further study should be done to verify our results by extending samples. |
