Objective:To observe the changes of L5 dorsal root ganglion in terms of tissue morphology, and the effect of acupotomy on IL-1,IL-6 and TNF-αin dorsal root ganglion of Rats with lumbar disc herniation, so as to make it clear that the acupotomy has the anti-inflammatory effect on the lumbar disc herniation and provide experimental support for the clinical treatment..Methods:Forty male SD rats were randomly divided into normal group, model group,electroacupuncture group and acupotomy group, four groups in all and 10 rats in each group. The incubation period of shrinkage claw of each group was tested by PL-200; The pathological changes of L5 dorsal root ganglion were observed by HE dyeing; Samples were took on eighth day after model, and the dorsal root ganglion were taken to observe the changes of IL-1, IL-6 and TNF-αby ELISA.Results:1. The changes of pain threshold and pathological changes of L5 dorsal root ganglion both suggest that lumbar disc herniation models were successfully constructed; acupotomy treatment on lumbar disc herniation can lighten the inflammatory response.2.The results of comparison of the IL-1 among all groups were as follows:comparing with normal group(10.01±0.55 ng/mg), the model group(14.15±1.51 pg/mg) increased significantly, and the difference had statistical significance (P<0.01); comparing with model group, electroacupuncture group (10.87±1.58 ng/mg) decreased significantly, and the difference had statistical significance (P<0.01); comparing with model group(12.19 ±1.15 ng/mL), acupotomy group(10.69±2.05 ng/mg) decreased significantly, and the difference had statistical significance (P<0.05).3.The results of comparison of the IL-6 among all groups were as follows:comparing with normal group(4.19±0.58 ng/g), the model group(5.54±0.74 ng/g) increased significantly, and the difference had statistical significance (P<0.01); comparing with model group, electroacupuncture group(4.74±0.82 ng/g) was no statistical difference(P>0.05); comparing with model group,acupotomy group(4.30±0.54 ng/g) decreased significantly, and the difference had statistical significance (P<0.01)4.The results of comparison of the TNF-αamong all groups were as follows:comparing with normal group(77.96±13.84 pg/g), the model group(112.94±11.17 pg/g) increased significantly, and the difference had statistical significance (P<0.01); comparing with model group, electroacupuncture group(84.06±6.27 pg/g) decreased significantly, and the difference had statistical significance (P< 0.01); comparing with model group,acupotomy group(81.54±5.60 pg/g) decreased significantly, and the difference had statistical significance (P<0.01)Conclusions:1. Acupotomology can effectively improve ultrastructura of Dorsal Root Ganglion with lumbar disc herniation and relieve inflammation lesion for Dorsal Root Ganglion.2. Acupotomy has a benign effect on regulating IL-1,IL-6 and TNF-a levels in Dorsal Root Ganglion with lumbar disc herniation.,this anti-inflammatory effect, which may be one of its mechanisms for lumbar intervertebral disc protrusion.
