Objective:The purpose of this experiment is to observe the change of the content of the substance P and CGRP in the dorsal root ganglion of the rat model,this model is the non-compressive animal model designed by Lu Zhidong.And the same time, we also observe the histological changes of the dorsal root ganglion.These obwervations will reveal the therapy mechanism of acupotomy nerve stimulation for disc herniation, provide the basis and a new train of thought for treatment of intervertebral disc herniation pain symptomsMethods:A total of 40 male Sprague-Dawley(SD) rats were randomly divided into four groups(n=10):blank group,model group,acupotomy group,and electroacupuncture group. The non-compressive model of experimental lumbar nucleus pulposus prolapse was built in model group,acupotomy group,and electroacupuncture group..Insert the rear disc,and take out the nucleus pulposus.The rats were punctured by injection of a solution containing autologous nucleus pulposus of coccygeal vertebral in the epidural cavnm.1 day after modeling, the acupotomy group and the electroacupuncture group were treated. After the treatment,the rats were opened the chest in a deep anesthesia and performed the heart perfusion.Then took the DRGs in L5 to test the contents of SP and CGRP of dorsal root ganglion by ELISA,and also the histological changes were observed.All groups of rats were test the hind limbs of thermal stimulation threshold before the modeling,after the modeling,and after the treatment respectively.Results:1 After the modeling, thermal hyperalgesia occurred in the hindlimbs of rats due to the nucleus pulposus in the epidural cavnm,and the hindpaw thermal withdrawal latencies were reduced.After the treatment of acupotomy, the hindpaw thermal withdrawal latencies of it were increased significantly(p<0.01).2The histological changes of DRG of the model group showed pathological characteristics typical of nerve roots.The neurocyte were shrinkage and had a disordered arrangement.The space between cells were widen and had vacuoloid changes.Histologically nissl bodies in nervous cells decreased.Some nucleus pycnosis and indistinct nucleoli also could be observed. After the intervention of acupotomy,the neurocyte arranged well and the nucleus were large and round.A few nucleus pycnosis and the intercellular spaces were smaller;after the intervention of electroacupuncture,a large number of nucleus pycnosis and few dissolved, The space between cells were widen and had a disordered arrangement of cells and nerve fibers.3The SP level of model group were increased significantly than the blank group(p <0.01),and the acupotomy of it were decreased than the model group(p<0.01);the conte nts of CGRP in the model group were higher than the blank group(p<0.05),and the ac upotomy of it were lower than the model group(p<0.05).Conclusion:1 Acupotomy nerve stimulation Can reduce model in rat dorsal root ganglion cells of pathological changes such as edema, degeneration, promote tissue repair and reduce inflammation in the ganglion,these may be one of the reason for pain relief.2Acupotomy nerve stimulation,by reducing the ganglion neural peptides:substance P, CGRP both pain conduction transmitter in content, makes the model rats of hyperalgesia and improving the pathological phenomena of pain facilitation, thereby reducing the generation of pain.
