Abstract:Objective:Study the influence of Qi Chai decoction on IL-4, IFN-y level in the serum of the model of allergic rhinitis rats, observe the pathological changes in nasal mucosa of the allergic rhinitis experimental rat model, and explore the role and efficacy in the model of allergic rhinitis rats.Methods:40healthy adult SD rats were randomly divided into five groups:normal control group (group A)8rats, the low-dose of Qi Chai decoction group (group B)8rats, the high dose of Qi Chai decoction group (group C)8rats, loratadine group (group D)8rats, allergic rhinitis group (group E)8rats. Rats with Allergic Rhinitis were made by taking Ovalbumin as a sensitizing agent. Corresponding drugs were given for14consecutive days in each experimental group after the cardiac blood collection, separation of the serum.Test the IL-4, IFN-y content using a double antibody sandwich ELISA, and observe the pathological changes in the rat nasal septum mucosa cells through HE staining experiments.Results(1)The nasal symptom score comparison before administration:each of the experimental rats nasal symptom score was significantly higher than the normal control group (0.87±0.83),which was statistically significant (P<0.01).(2) The nasal symptom score comparison after administration:The model group nasal symptom score (6.62±1.06)) is significantly higher than the normal group (1.00±0.75),which was statistically significant (P<0.01); Compared with the model group,the nasal symptom score of the low dose of Qi Chai decoction group (3.12±0.99),the high dose of Qi Chai decoction group(1.75±1.13)and the desloratadine given group (1.25±1.03) are significantly lower,there are significant differences(P<0.01); There is no significant difference between the nasal symptom scores of the Qi Chai decoction high dose group and the scores of the normal control group (P>0.05).(3) The count of model group nasal mucosa eosinophil (27.50±2.39) is obviously more than the normal control group (2.25±1.03),there are significant differences(P<0.01); Compared with the model group,The count of eosinophil in the low dose of Qi Cai decoction group(6.25±1.67),the high dose of Qi Chai decoction group(4.25±1.03)and the desloratadine given group (4.50±1.06)are significantly lower, there are significant differences (P<0.01); comparing the eosinophil count in the low-dose group of Qi Chai decoction and the high-dose group of Qi Chai decoction, the difference was statistically significant (P<0.01).(4) The serum IL-4levels of the modle group(262.64±32.11), are higher than the normal group (55.34±15.77), there is a significant difference (P<0.01); Compared with the model group,The serum IL-4levels of the low dose of Qi Chai decoction group (218.65±45.03),the high dose of Qi Chai decoction group(136.36±42.22)and the desloratadine given group (173.44±49.47) are significantly lower, there are significant differences(P<0.01, P <0.05); IL-4level of the low dose of Qi Chai decoction group was higher than the high dose of Qi Chai decoction group, the difference was statistically significant (P<0.05).(5) The serum IFN-y levels of the modle group(38.40±12.58) are lower than the normal group (148.99±14.80), there is a significant difference (P<0.01); Compared with the model group,the serum IFN-y level of of the low dose of Qi Chai decoction group(58.67±20.12),the high dose of Qi Chai decoction group(127.90±15.68), and the desloratadine given group (106.98±18.58)are significantly higher, there are significant differences(P<0.01, P <0.05); Compared with the the high dose Qi Chai decoction group,the serum IFN-y level of the low dose Qi Chai decoction group and loratadine given group are lower, the difference is statistically significant (P<0.05).Conclusion:(1)The replication of allergic rhinitis (AR) rat model is successful.(2) The Qi Chai decoction can improve nasal symptoms of allergic rhinitis in rats,can improve nasal mucosa inflammatory infiltration,and the high-dose group was better than the low-dose group.(3) the Qi Chai decoction can reduce the serum IL-4levels of the rats with allergic rhinitis.(4) the Qi Chai decoction can raise the serum IFN-ylevels of the rats with allergic rhinitis.(5) The mechanism of the Qi Chai decoction prevention and treatment to the rats with allergic rhinitis may relate to reducing serum IL-4levels and increasing IFN-y levels.
