| Objective:Through observing the rat eye symptoms and signs of change of experimental autoimmune uveitis (EAU), with Tumor Necrosis Factor-a (TNF-a), Interleukin-6 (IL-6), Interleukin-1 (IL-10) as observation indexes, objectively evaluate the therapeutic effect of Qufeng Huoxue soup, and to explore the development of TNF-a, IL-6, IL-10 in action during the uveitis.Methods:1. Separated and extracted of bovine retinal S antigen; 2. The EAU model:25 SD rat of the SPF level, the two foot pads was injected of bovine retinal S antigen to establish the model of experimental autoimmune uveitis, blank group with 5 rats, injection of physiological saline.3. Cure with Qufeng Huoxue soup:The 20 successful modeling rat were divided into 4 groups, namely control group, the high dose group (4g/Kg.d), the middle dose group (3g/Kg.d), the low dose group(2g/Kg.d), Were given Qufeng Huoxue soup 4g/Kg.d,3g/Kg.d,2g/Kg.d, the control group and blank group were given normal saline treatment, observed the expression of rats’ eyes everyday.4.After 28 days, all the treatment rats were killed, sacrificed effects of the peripheral side by ELISA test for TNF-a, IL-6, IL-10.Results:1. Successfully extracted and measured the bovine retinal S antigen.2. The EAU model was successfully induced in SD rats, and induced a success rate of 80%.3. With Qufeng Huoxue soup for 28 days, test of the contents of the rats’ serum TNF-a, IL-6, IL-10, the control group was significantly higher than the other groups. There was no significant difference between the control group and high dose group, so was the low dose group and middle dose group. The remaining were statistically significant between groups.Conclusion:1. The EAU model Can be induced with bovine S antigen injection.2. The treatment of Qufeng Huoxue soup with EAU was effective, and the effect of high dose group was higher than that in low dose group and middle dose group. |
