Surface Plasmon Resonance (SPR) technology has been becoming a leading one for biomolecular interaction analysis because of its specific advantages. Biosensors based on SPR technology have been widely used in various fields such as biology, medicine, chemistry, and so on. More and more scientists devote themselves to researching SPR technology and its applications.This thesis reviews the research evolvement of biosensors based on SPR technology in the world, demonstrates the SPR principle theoretically, analyzes specialities of protein molecules and their interactions briefly. Based on these, methods to realize detecting interactions between protein molecules by SPR technology are studied seriously, various factors to effect biosensors based on SPR technology are discussed deeply, and the effecting results are simulated numerically by matlab tools.A biosensor based on SPR technology is presented which has been manufactured by ourselves. The concept of virtual apparatus was introduced into the design process, to shorten the time completing the biosensor's system, to adapt the biosensor to multipurpose measuring, and to be convenient for developing this system later; the movable parts of the biosensor were reduced by increasing the convergence angle of incident light to improve the biosensor's veracity; the structure of sensing parts was improved and a microfluid system was introduced to adapt the biosensor to realtime detecting interactions between protein molecules.The biosensor system has been debugged and the problems which were found have been solved. At last, some new ideas about studying biosensors based on SPR technology in the future are presented.
