Aims:Postmenopausal women, due to the decline of ovarian function, estrogen secreted by the ovaries will greatly reduce,the phenomenon will result in the incidence of obesity, it has been confirmed by experiments. Obesity will often easily lead to the occurrence of some diseases, such as:coronary heart disease, insulin resistance and so on. it is necessary to take certain measures to alleviate the symptoms of obesity. Postmenopausal women at home and abroad simulation model is commonly used in bilateral ovariectomized rat model, after ovariectomy in mice, decreased estrogen secretion, symptoms easily lead to obesity in animals. Exercise therapy is one of the most effective way to prevent of obesity, aerobic exercise is the preferred movement pattern of obesity prevention and treatment. In this study, according to observation of the effect of treadmill exercise on ovariectomized mice serum lipids, adipocyte morphology and cytokine gene expression,the Exercise undertook preliminary research and discussion on preventing obesity after menopause.Methods:The 24 8-week old female C57 mice were divided into three groups, which were randomly divided into a quiet group (SHAM), a quiet group of ovariectomized (OVX), ovariectomized group and run uphill (OVX+UP), n= 8. Ovariectomized mice were anesthetized surgery, SHAM group were removed a little fat around the ovaries. Training period lasts 8 weeks. After 8 weeks of training,the mice were sacrificed by removal of the eyeball method, collection of blood serum,then separated the blood serum for testing relatively lipid-related indicators, such as:triglycerides, free fatty acids and total cholesterol levels; take perirenal white adipose tissue in mice, in part for the staining to observe perirenal white fat cells in vivo morphological changes; part of them for the detection of perirenal adipose tissue adipokines:adiponectin, PPARγand C/EBP-αmRNA expression. Finally, three sets of experimental results are compared and analyzed to verify whether exercise can affect body fat content. Results:(1) The comparison of results of body weight and food intake:The comparison of results of body weight:the OVX group's body weight is the highest, the OVX+UP group followed, the SHAM group's body weight is the lowest, and there are significant differences among the groups; The comparison of results of food inta-ke:between the OVX+UP group and OVX group,there is a significant differ- rence, between SHAM group and the OVX group there is a significant differ- ence, between the OVX+UP group and the SHAM group have no significant differrrrence.(2) The comparison of results of serum lipid levels and adipocyte morphological changes:The comparison of results of serum total cholesterol level, between OVX group and the SHAM group, which have highly significant difference, between the OVX+UP group and the OVX group, which have highly significant differences, and other groups show no significant difference; The comparison of results of free fatty acid levels, between OVX group and the SHAM group, which have highly significant differences, the other groups show no significant difference; The comparison of results of triglyceride levels, between the OVX+UP group and the SHAM group, which have highly significant differences, between the OVX+UP group and the OVX group, which have highly significant differences, other groups no significant difference. The comparison of results of adipocyte morphological changes,the OVX group's cell size is the largest, cell number is the least at the same field of vision; OVX+UP group, the cell number and volume of medium, between the SHAM group and the OVX groups; SHAM group is the minimum volume of fat cells, and the largest number of fat cells.(3) The comparison of results of adipocyte gene expression comparison results:The comparison of results of adiponectin gene expression:between the OVX+UP group and the OVX group, which have significant differences, between the OVX+UP group and the SHAM group, which have significant differences, between the SHAM group and the OVX group, which have significant differences; PPARγgene expression results show that: between the SHAM group and the OVX group,there is a significant difference, between the OVX+UP group and the OVX group have not significant difference, between the OVX+UP group and the SHAM group were not significantly different; The C/EBPαgene expression results showed that:the OVX+ UP group and SHAM group have highly significant Differences, SHAM group and the OVX group compared to a highly significant difference, between the OVX+UP group and the OVX group, did not find significant differences.Conclusions:(1) Combined with the amount of mice body weight and dietary changes, we can obtain that, the mice eating the amount of difference has little effect in changing of their body weight, increased body weight mainly due to ovariectomy.And uphill running aerobic endurance training can ease body weight which was increased by being ovariectomized.(2) it can be directly observed from the fat cells morphological,ovariectomy can result in increased fat cell size, and aerobic endurance training can reduce the size of fat cells, thus effectively improving obese.(3)The degree of obesity can increase and the fat content increased, so that the relevant lipid parameters were significant changes, and uphill running aerobic exercise can effectively alleviate increase blood lipid levels, Alleviated to some extent the adverse effects of obesitye.(4)Ovariectomy can make mice' associated cytokines significant changes occur, lead to obesity, and aerobic endurance training can regulate the related gene expression levels to effectively improve the symptoms of obesity.
