Effects Of Connexin26 And Connecxin43 On The Proliferation And Invasiveness Of Bladder Transitional Cell Carcinoma

[Objectives] To build the plasmid of Cx26 and Cx43 gene and transfer them into T24 Bladder Transitional Cell Carcinoma (BTCC), discuss the effect on invading and multiplication ability of BTCC while the Cx26 and Cx43 express.[Methods] Use the primer premier software designs primers, redouble Cx26 and Cx43 gene by PCR methods and identify that by electrophoretic analysis. Restriction enzyme double digests eukaryotic expression vector pEGFP-N3,Cx26 and Cx43 and then make the Cx26 and Cx43 gene insert the polyclone locus of pEGFP-N3 . Transfer the plasmid of Cx26 and Cx43 gene into T24 BTCC cell ,select cell lines after cultured by LB medium ,identify expressing the protein of Cx26 and Cx43 successfully By western blot and green fluorescent, then use this cell make the artificial basement membrane invasion test and proliferation test. The ability of invasion is denoted By counting the cells that penetrate artificial basement membrane, the cell proliferation activity is indicated by MTT test.[Results] 1. Build the plasmid of Cx26 and Cx43 gene successfully; 2.comparing with the cell line that the group of control and transferred pEGFP-N3 only, the ability of invasion decreased observably in cell line that the group of transferred Cx26 or Cx43 gene; 3. The MTT test showed that comparing with the cell line that the group of control and transferred pEGFP-N3 only, the multiplication capacity decreased significantly in cell line that the group of transferred Cx26 or Cx43 gene.[Conclusion] The plasmid of Cx26 and Cx43 gene can be built, the expression of Cx26 or Cx43 gene may have the effect that inhibit the BTCC cell invasion and proliferation.
