Objective:GRK6 and Connexin26(Cx26)are involved in cell communication.GRK6 is an important serine/threonine protein kinase in the human body belonging to the GRK superfamily,which participate in signal transduction regulation and control between cells and cells of the body.While the Cx26 can be widely expressed in a variety of human tissues and epithelial cells and mainly take part in regulating cell proliferation,differentiation,metabolism in body and other life activities as the basic composition unit of gap junction channel protein which is involved in the material exchange and transmission function in cells.But the expression and effect research report of them are less reported in the gastric mucosa lesions of different pathological changes including in the normal gastric mucosa,atypical hyperplasia in varying degrees and different types of gastric cancer cell expression at present.As the study of gastric cancer and precancerous lesions of cells found in recent years,the former is associated with poor prognosis of tumor cells such as poorly differentiated,infiltration,and metastasis,whereas the highly expression of the latter in the cytoplasm affects the occurrence and development of gastric cancer cells and may have a certain inhibitory effect on tumor progression.The prognosis of patients with gastric cancer is closely related to tumor differentiation degree,infiltration depth,lymph node and blood line transfer,and so on.In order to further improve the gastric cancer patients'survival time and improve their quality of life,we can delve into the possible molecular biology mechanisms of different types of gastric cancer and precancerous lesions,find a reliable and efficient measure of molecular biology for the early diagnosis and evaluation of prognosis,and look for new potential targets of treatment and monitoring prognosis.Through the detection of GRK6 and Cx26 expression in gastric carcinoma,corresponding normal tissue adjacent to it and atypical hyperplasia of different level in gastric mucosa tissue cells,this experiment analysis their expression differences between different tissue cells,explore their expression and changing rules with the progress of the cell types in the corresponding organization,investigate the correlation and the possible mechanism of their expression in respective tissues and cells,and discuss the clinical value of GRK6 and Cx26 in the early diagnosis of gastric cancer,targeted therapy and prognosis evaluation.Methods:1.In this study,we randomly selected 169 archival paraffin blocks from Department of Pathology,the first affiliated hospital of the Southwest Medical University,including surgical excision specimens and digestive endoscopic biopsy tissue samples from 2013 to 2014.82 cases of gastric carcinoma,52 cases of atypical hyperplasia,and 35 cases of normal gastric mucosa(from the edge of the tumor more than 5cm)are involved.We detected the expression of GRK6 and Cx26by Immunohistochemical Envision method and analyzed the relationship in each groups.2.Results were marked according to Shimizu scoring method.According to each section of the cells positive staining degree score.3.SPSS21.0 statistical software was used for statistical analysis,from which spearrman correlation analysis,fisher exact probability calculation method and the chi-square test were mainly adopted.Test for the P<0.05 was considered statistically significant.Results:1.The positive expression rate of GRK6 in normal gastric mucosa,mild-to-moderate dysplasia,severe atypical hyperplasia,intestinal type gastric cancer group,and diffuse gastric cancer group were respectively 0%,0%,3.3%,41.5%,65.5%.In 82 cases of gastric carcinoma,the positive expression number of GRK6 were 41 cases,and the expression rate was 50.0%.The positive expression number of GRK6 in normal tissue adjacent to carcinoma is 0(x~2=12.761,P=0.000).The positive expression number of GRK6 in atypical hyperplasia tissues was only 1 case,and the expression rate was 3.3%(x~2=28.228,P=0.000).GRK6 expression levels of normal gastric mucosa and dysplasia group was significantly lower than the expression level of gastric cancer(P<0.05),statistically significant.2.The positive expression rate of Cx26 in normal gastric mucosa,mild-to-moderate dysplasia,severe atypical hyperplasia,intestinal type gastric cancer group,and diffuse gastric cancer group were respectively 2.9%?4.6%?10.0%?47.2%?24.1%.In 82 cases of gastric carcinoma,the positive expression number of Cx26 were 32 cases,and the expression rate was 39.0%,of which the positive expression rate of intestinal type gastric cancer was 47.2%,and the positive expression rate of diffuse gastric cancer was 24.1%.Cx26 expression level of diffuse gastric cancer was significantly lower than that of intestinal gastric cancer(x2=4.179,P=0.041).The positive expression number of Cx26 in normal tissue adjacent to carcinoma was 1 case,and the expression rate was 2.9%(x2=16.718,P=0.000).The positive expression number of Cx26 in Atypical hyperplasia tissues were 4 cases,and the expression rate was 7.7%(x2=10.182,P=0.001).Cx26 expression level of normal gastric mucosa and dysplasia group was significantly lower than the expression level of gastric cancer(P<0.05),statistically significant.3.GRK6 and Cx26 expression had no significant correlation with patients'gender and age(P>0.05),but related to tumor types.Positive expression rate of intestinal gastric cancer was significantly higher than that of diffuse gastric cancer.Conclusion:1.The positive expression of GRK6 in gastric cancer was high.As the tumor infiltration depth,lymph node metastasis and pathologic stage increased,its expression level increased gradually,which prompted GRK6 may participate in gastric cancer invasion and metastasis.2.Cx26 in gastric cancer was increased compared with that in normal gastric mucosa accompanied by the change of expression site,and had a close correlation with the depth of tumor invasion,the degree of differentiation and lymph node metastasis.It suggested that CX26 inhibited progression and metastasis of gastric cancer,which may be related to its promoting cell differentiation.3.GRK6 and Cx26 expression was negatively correlated in gastric cancer cells,suggesting that Cx26 may inhibit the expression of GRK6,then inhibiting tumor invasion and metastasis.4.Through testing and analyzing GRK6 and Cx26 expression in gastric cancer tissue,we make new detection of gastric cancer metastasis and biological indicators of prognosis evaluation possible,provide further research for effective and feasible gastric cancer targeted drugs,and offer a new thought and method of diagnosis and treatment for gastric cancer patients.
