Objective:In this study,we will investigate the process of the vascular remodeling induced by vascular cell phenotype transformation and cell proliferation involved in spontaneously hypertensive rats.We also demonstrate that Ramipril alleviates cerebral vascular remodeling in spontaneously hypertensive rats by inhibiting the process of vascular cell phenotype transformation and cell proliferation.It preliminarily revealed that the protective mechanism of Ramipril for the cerebral vascular remodeling induced by vascular cell phenotype transformation and cell proliferation in spontaneously hypertensive rats,which provides a new theoretical basis for the treatment of spontaneously hypertensive rats in the future.Methods:In this study we used healthy adult WKY and SHR.According to the random principle,the experimental rats were divided into four groups: WKY group,WKY + Ramipril group,SHR group,SHR + Ramipril group.Systolic blood pressure and diastolic blood pressure were measured by non-invasive tail artery manometry.HE staining was used to detect the morphological changes of brain arteries in each group.The systolic function of cerebral arteries in each group was detected by pressure maneuver method.The expression and distribution of Cx43 in each group were observed by immunofluorescence and immunohistochemistry.The expression of Cx43 mRNA in each group was detected by RT-PCR.Blot technology detects protein expression of Cx43,PCNA,?-SMA and OPN.Results:(1)Compared with the WKY group,the systolic and diastolic blood pressures of SHR rats were significantly increased,and the cerebral vascular injury was severe;cerebrovascular cells Cx43,cerebral vascular cell contractile marker protein(OPN)and The expression of cell proliferation related factors(PCNA)was significantly increased,and the expression of cerebral vascular cell secretory marker protein(?-SMA)was significantly decreased;the cerebral vasoconstriction function was abnormally increased.(2)After the intervention of Ramipril,the systolic blood pressure and diastolic blood pressure of SHR + Ramipril group decreased significantly,the vascular remodeling was relieved;the expression of Cx43,OPN and PCNA were decreased,and the expression of ?-SMA was increased;the cerebral artery vasoconstriction function was significant decline.Conclusion:(1)The vascular remodeling mediated by vascular cell phenotype transformation and cell proliferation was involved in the process of spontaneously hypertensive.(2)Ramipril protects the cerebral artery injury after spontaneously hypertensive rats,which may inhibit vascular cell phenotype transformation and cell proliferation and improve the cerebral artery vascular remodeling.
