Objective:To observation of curcumin (curcumin, CUR) on rat traumatic cataract (traumatic cataract, TA) lens epithelial cells (lens epithelial cells, LECs) of nuclear factor-κB (nuclear factor-kappa B, NF-κB) and cyclooxygenase-2 (cyclooxygenase-2, COX-2) expression and activation of the lens epithelial cell apoptosis.Methods: The 100 male SD rats were randomly assigned according to the principle of divided into five groups, each with 20, that is, the blank control group, negative control group, positive control group, low concentrations of CUR group, high concentrations of CUR group. Establishment of rat model of traumatic cataract, the effects of different concentrations of curcumin for different time points (12h, 24h, 72h, 168h) the impact of the formation of rat lens opacity, was observed under the microscope lens epithelial cells and lens fiber structure changes. By immunohistochemical SABC staining at different times (12h, 24h, 72h, 168h) lens epithelial cells, NF-κB and COX-2 expression in the case, by using terminal deoxynucleotidyl transferase-mediated nick end labeling ( terminal deoxynucleotidyl transferase mediated dUTP-biotin nick end labeling, TUNEL) detection of lens epithelial cell apoptosis cells, and the associated comparative statistical analysis.Results:1. Using the anterior capsule cut open law can be successfully established rat model of traumatic cataract.2. In the normal control group, the rat lens has to be transparent state; while in traumatic cataract model group, 12 hours after the lens appear cloudy, 24-hour lens opacity increased slowly, 72 hours to accelerate the progress of lens opacification, 168 hours a totally lens opacity. Lens opacities indications over time, to extend the CUR group modeling lens turbidity significantly lighter than the control group (P<0.01). 3. In the light microscope, by HE staining found in experimental lens capsule wound not see the normal cuboidal epithelium, epithelial cells, irregular hyperplasia, the formation of new lens capsule to wound healing, traumatic lens fibers channel disorder.4. Immunohistochemistry revealed negative control group of lens epithelial cells NF-κB activation and COX-2 expression was significantly higher compared with the control group, differences were statistically significant (P<0.01). And the modeling of time with the extension of LECs in the negative control and NF-κB activation of COX-2 expression gradually increased, the time difference between the positive expression rate was statistically significant (P<0.01).5. CUR can inhibit the NF-κB activation and COX-2 expression and reduce the degree of lens opacity, and there is a certain degree of dose-dependent.6. TUNEL detected, CUR group of all time apoptosis rate were significantly lower than the control group (P<0.01).Conclusion:1.Traumatic cataract, NF-κB, and COX-2 expression, on the development of cataract play an important role.2.Curcumin in rats with traumatic cataract have anti-inflammatory and anti-apoptosis.3.Curcumin anti-inflammatory and anti-apoptotic mechanism may be related to inhibiting NF-κB and the activation of COX-2 expression.4.Curcumin has a certain inhibition of early mild traumatic cataract formation.
