Objective:The discussion about the effect of curcumin on anti-oxidative damage in traumatic cataract mechanism.Methods:102 disease-free rats for SD rats were used and divided randomly: 20 of SD rats were used for the normal control group; the other of 80 were cut anterior capsule to make as traumatic cataract model, and then randomly divided into traumatic cataract model group, high strength of curcumin treatment group, medium strength of curcumin treatment group and low strength of curcumin treatment group. Used slit-lamp to observe the groups of rats and recorded the changing process of lens turbidity. In 1st, 3rd, 7th, 14th days, kill lens respectively, the lens were separated rapid, one of lens from each group will be made to paraffin section. Changes in structure of Lens epithelial cells (LECs) were observed using HE staining. The rest lens were be observed by visible light Spectrometry about the activity of glutathione peroxidase (Glutathion peroxidase, GSH-PX), peroxide catalase (Catalase, CAT), superoxide dismutase (Supero-xide dismutase, SOD), malondialdehyde (Melondialdehyde, MDA) in the organization, and data statistics and analysis.Results:1. Lens transparency: there is no change about transparency in the normal control group;but medium and high concentrations of curcumin treatment group was significantly higher than traumatic cataract group and low concentration of drug treatment group which has outstanding statistical significance (P<0.05). 2. In the blank control group, the structure of lens is essentially normal under optical microscope; in the model control group, damage to lens structure appeared in different various concentration drugs. 3. Antioxidant enzyme activity: the activity of GSH-PX, CAT, SOD in Traumatic cataract group significantly lower than the normal control group, the activity of MDA was significantly higher (P<0.05); for GSH-PX, CAT, SOD, medium and high concentrations of curcumin treatment group with traumatic cataract model group low concentration of drug treatment group was significantly increased (P<0.05), MDA activity decreased significantly(P <0.05).Low-concentration treatment group and model group, no statistically significant difference(P>0.05).Conclusions:1. needle penetration method can be more successful in making traumatic cataract rat model.2. oxidative damage is the important factor which led to traumatic cataract.3. curcumin in moderate and high concentrations (0.1mg/0.1ml,0.2mg/0.1ml) can keep lens activities of antioxidase and restraining oxidative damage effect obviously which can effective control and delay early stage of traumatic cataracts.
