Objective:Through the tissue chip technique, the level of y-aminobutyric acid A(GABAA) receptorα1 subunit in human brain was examined by immunohistochemistry, and applying the three dimensional reconstruction technique, its position state was reconstructed according to examination value to gain the three-dimensional distribution map of GABAA receptorα1 subunit in brain. So we can study the relationship between the dissection structure and the material distribution, and study the mechanism of benzodiazepine's poisoning and depedence, and provide experimental evidence or theoretical basis. Meanwhile three-dimensional reconstruction would also be used for medical research.Methods:The collection postmortem examination's human brain specimen (lifetime physical health,brain structure is complete, and non-naked eye pathological change), The entire brain was cut into 5×5×5mm scraps according to the Cartesian coordinate slivers. All the scraps were selected, and their coordinate figures were recorded. The tissue chip was produced after making the conventional paraffin blocks. Then the GABAAα1 acceptors were examed by immunohistochemistry, and the results were statistical analysised using the SPSS software. So we can know the relationship between the distribution of GABAA receptorα1 subunit and anatomical structure. Then we established the three-dimensional perspective through three-dimensional reconstruction technology, and obtained the direct-viewing material distribution model.Results:1.GABAA receptorsα1 subunit's expression in various brain regions was not exactly the same (P<0.05), and the descending order from high to low was:hippocampus, cerebral cortex, striatum, thalamus, and brain stem. The white matter almost had no expression. 2.The expressions of the GABAA acceptorα1 subunit in different parts of cerebral cortex had no difference (P>0.05);3.The three-dimensional distribution of GABAA receptorsα1 subunit in brain was successfully constructed.Conclusion:1.The descending order of the GABAA receptorsα1 subunits in human brain from high to low is:hippocampus, cerebral cortex, striatum, thalamus, and brain stem.2.The different anatomical structure of GABAA receptorα1 subunit in the brain suggests that the benzodiazepine works in the corresponding position, which was consistent with the existing research.3.The three-dimensional reconstruction is help to know the dimensional relationship between the regional structure and distribution, and the observation of drug action of benzodiazepine in the corresponding location.
