| Objective:This study was designed to investigate expression of N-acetylcholine receptor in the adult's brain by tissue microarray with immunohistochemical techniques.To compare it in different regions and find out the relationship between receptor's distribution and anatomical structure in the brain. Also reconstruct the model of the receptor's distribution by computer three-dimensional reconstruction graphics.Methods:The brain specimens were collected which have complete structure and no visible lesions in autopsy. Cut it into small pieces(5mm*5mm*5mm) by position select instrument, take the whole brain and record the coordinates of every pieces. Make it to common paraffin blocks and then to tissue microarrays and slices. Stain it by Immunohistochemical method use rabbit anti-human N-acetylcholine receptor monoclonal antibody. Detect the expression and distribution of N-acetylcholine receptors in the human brain. Analysis the data by SPSS statistical software, reconstruct the distribution of N-acetylcholine receptor to 3D by VTK software, get a model aboult the distribution of N-acetylcholine receptor and then visualize it.Result:Compare the expression level of N-acetylcholine receptor in various regions of the brain (P<0.05), the expression level is not exactly the same, then compare each other by pairwise comparison (P<0.05), the hippocampus showed the highest expression level. The corpus striatum and cerebral cortex showed the second highest expression level. The expression intensity was hugely different from those in some other regions. We compared the expression of N-acetylcholine receptors by pairwise comparison for the same regions of both sexes (P>0.05), the expression intensity has no difference.Conclusion:N-acetylcholine receptors have widely expressed in human brain, and show a regional distribution.The hippocampus, the corpus striatum and cerebral cortex showed high expression level relating to the functions of study, memory and cognitive skill.The expression intensity of N-acetylcholine receptors have no significant difference between males and females. |
