Objective: To study the effect of electro-acupuncture "Quchi" and "Yanglingquan" combined with Jiawei Shaoyao Glycyrrhiza decoction on the expression of GABAa receptor and BDNF,SYN in hippocampus of rats with stroke limb spasm,and to explore the mechanism of acupuncture and medicine combined with acupuncture and medicine in the treatment of stroke limb spasm.Methods: According to the random digital table,63 SD rats were randomly divided into blank group,pseudo-operation group,model group,electro-acupuncture group(electroacupuncture Quchi,Yanglingquan),traditional Chinese medicine group(Jiawei Shaoyao Glycyrrhiza decoction decoction),acupuncture and medicine combination group(EA Quchi,Yanglingquan administration Jiawei Shaoyao Glycyrrhiza decoction)and western medicine group(Ba-chlofen solution)with 9 rats in each group.The model of stroke limb spasm was established by injecting NMDA into the internal capsule of the model group,and each treatment group b-egan to give the corresponding treatment method once a day for 5 days from the 1st day after the successful establishment of the model,the rat model of stroke limb spasm was established by injecting NMDA into the internal capsule of the modified NMDA thread thrombus method.Except for the blank group,all the groups were bound and bound with the acupuncture gr-oup.The rats in the pseudo-operation group,the model group and the acupuncture group were given the same amount of saline for 5 days.The neurological function and muscle tension score of rats in each group were observed after modeling and treatment,the ratio of type I and type II muscle fibers in left biceps brachii and quadriceps femoris was analyzed by skeletal myosin ATP staining,the microsuperstructure of hippocampal synapses was observed by ultrastructure transmission electron microscope,and the gene and protein expression of GABAa receptor and BDNF,SYN in hippocampus were detected by RT-PCR and WB,respectively.Results:1.Zea longa score: compared with the blank group,False operation group was no significant difference(P>0.05),and the score of the model group was significantly higher than that of the blank group(P<0.01),suggesting that the model was successful.Compared with the model group,the scores of each treatment group were lower and statistically significant(P<0.05),and there was no difference among the three groups(P>0.05).The results showed that electro-acupuncture,traditional Chinese medicine,acupuncture and medicine combination and western medicine could improve the muscle tension of rats,but there was no significant difference among the three groups.2.Modified Ashworth muscle tension evaluation: compared with the blank group,False operation group was no significant difference(P>0.05),and the score of the model group was significantly higher than that of the blank group(P<0.01),suggesting that the model was successful.Compared with the model group,the scores of each treatment group were lower and statistically significant(P<0.05),and there was no difference among the three groups.The results showed that electro-acupuncture,traditional Chinese medicine,acupuncture and medicine combination and western medicine could improve the muscle tension of rats,but there was no significant difference among the three groups.3.The proportion of type I and type II muscle fibers in left biceps brachii and quadriceps femoris in rats: there was no significant difference in the proportion of type I muscle fibers in left biceps brachii and quadriceps femoris in each group(P > 0.05).Under the condition of alkali dyeing(PH10.7),type I fiber showed light gray and type II fiber showed dark brown.The results of staining showed that the muscles of each group were dissolved to varying degrees,and there was no chess-disk structure.The distribution pattern of type I and II fibers changed and the fiber spacing increased.4.The expression of GABAa receptor,BDNF and SYN gene in the hippocampus of rats: the model group was significantly lower than that of blank group and false operation group(P<0.01),Compared with the model group,the expression of each treatment group increased significantly(P<0.05,P<0.01);Compared with traditional Chinese medicine group,The expression of acupuncture and Chinese medicine combination group and western medicine group increased significantly(P<0.05,P<0.01);Compared with acupuncture group,The expression of acupuncture and Chinese medicine combination group and western medicine group was significantly higher than that of acupuncture group(P<0.05,P<0.01).5.The expression of GABAa receptor,BDNF,SYN protein in hippocampus of rats :the model group was significantly lower than that of blank group and false operation group(P<0.01),Compared with the model group,the expression of each treatment group increased significantly(P<0.05,P<0.01);Compared with traditional Chinese medicine group,The expression of acupuncture and Chinese medicine combination group and western medicine group increased significantly(P<0.05,P<0.01);Compared with acupuncture group,The expression of acupuncture and Chinese medicine combination group and western medicine group was significantly higher than that of acupuncture group(P<0.05,P<0.01).Conclusion:1.Acupuncture of Quchi,Yanglingquan combined with Jiawei Shaoyao Glycyrrhiza decoction can improve the nerve function and muscle tension of stroke limb spasm.2.The combination of acupuncture and medicinecan effectively improve the spasm of stroke limbs.The mechanism may be achieved by up-regulating the gene and protein expression of GABAa receptor,BDNF,SYN in hippocampus of rats.
