Glaucoma filtration surgery is a conventional operation; the successful rat of operation is closely related to the fibrosis in drainage bypass. Using anti- metabolism drugs such as mitomycin-C (MMC),5-fluorouracil (5-Fu)can improve successful rate of the surgery, but severe undesirable side effects like hypotonous maculopathy, wound leakage, and bleb-related endophthalmitis are unavoidable. To maintain the filtering bleb without or with fewer complications, various physical barriers, chemical and biologic agents have been applicated after filtration surgery, and have achieved a good therapeutic effect. However, due to various reasons, they have not yet widely used in clinical practice.Ski gene is a proto-oncogene, as a transcription coactivator or corepressor in vivo regulating the expression of a number of important transcription factors, and plays an important role in the wound tissue regeneration. Our preliminary study found that, C-ski gene can promote rat skin fibroblasts proliferating and reduce collagen secretion in vitro, and also can promote wound healing and reduce scar on the rabbit wounding ears, so we speculated that C-ski gene may be can promote wound healing, reduce scar formation. We will use it after trabeculectomy in rabbit eyes to observe the effect, in order to lay the foundation for future clinical application.The study is divided into three parts: 1. Recombinant pcDNA3.0 plasmid containing C-ski gene and naked plasmid pcDNA3.0 be amplified, purified, and identified. 2. Different dose C-ski gene influence scar formation after trabeculectomy in rabbits. 3. 500ug recombinant pcDNA3.0 plasmid containing C-ski gene, PBS, naked plasmid pcDNA3.0, and Mitomycin-C (MMC) influence scar formation after trabeculectomy in rabbits.The main results and conclusions are as follows:1. The recombinant pcDNA3.0 plasmid containing C-ski gene and naked plasmid pcDNA3.0 are amplified, purified, and identified. The results confirm that: We have already collected the recombinant pcDNA3.0 plasmid containing C-ski gene and naked plasmid pcDNA3.0 by alkaline lysis method, the dose and quality were enough to satisfy our animal experiments requirements.2. Different doses of recombinant pcDNA3.0 plasmid containing C-ski can reduce the intraocular pressure and extend the bleb survival time, and can inhibit the typeâ… collagen production in drainage bypass, as the dose increased, the effect enhanced, while fibroblasts and myofibroblasts generation and transformation had no significant effect.3. The application with 500ug doses of recombinant pcDNA3.0 plasmid containing C-ski after trabeculectomy in rabbit eyes ,did not cause inflammation, can significantly reduce the typeâ… collagen production, inhibit the formation of scar to extend the time of postoperaton low intraocular pressure, without mitomycin C (MMC) side-effects.
