Objective:To study the histopathologic changes of filtering blebs after applying BAPN after trabeculectomy in rabbit eyes.Methods:48 healthy conspecific New Zealand white rabbits(48 right eyes)were randomly divided into A,B,C,D 4 groups(12 eyes in each group)?Group A underwent trabeculectomy only;group B underwent trabeculectomy with 0.2mg/ml BAPN cotton patch placed under sclera flap for 3 minutes;group C underwent trabeculectomy with 1.0mg/ml BAPN cotton patch placed under sclera flap for 3 minutes;group D underwent trabeculectomy with 5.0mg/ml BAPN cotton patch placed under sclera flap for 3 minutes.Side effects and complications were evaluated with slit-lamp,and histopathological changes were investigated with HE?Masson staining and immunohistochemistry at 14 days,28 days after trabeculectomy in rabbit eyes?Results: No significant statistical differences were found in anterior chamber inflammation with slit-lamp among each group(P>0.05).After 14 days,there were significant statistical differences in the number of fibroblasts between group A and group C(P <0.05);there was no significant statistical difference in the amount of collagen fibers in group A,B,C and D(P> 0.05);there was no significant statistical difference in the number of TGF-?1 positive cells between group A and group B(P> 0.05);the number of TGF-?1 positive cells in group A and group B was significantly different(P <0.05).After 28 days,there were significant statistical differences in the number of fibroblasts between group A and group C(P <0.05);there were significant statistical differences in the amount of collagen fibers between group A and group D(P <0.05);there was no significant statistical difference in the number of TGF-?1 positive cells between between group A and group B(P> 0.05);there were statistical significant differences in the number of TGF-?1 positive cells in the remaining groups(P <0.05).Conclusions:Trabeculectomy combined with BAPN can inhibit the proliferation of fibroblasts and the proliferation of collagen fibers after operation,inhibit or reduce the postoperative filter scarring changes,,and The inhibitory effect is related to the concentration of BAPN and the time of action.
