Objective:To differentiate excessive internal heat of liver syndrome and Yin-deficiency excessive Yang syndrome of hypertension by analyzing the differences based on metabolomics approaches. Groups from excessive internal heat of liver syndrome of hypertension and Yin-deficiency excessive Yang syndrome of hypertension were screened, evaluated and marked to explore the basic molecular substances of TCM syndrome on hypertension, while supplying more clinical trial evidences about TCM syndrome and providing supports for differentiation of TCM syndrome types on hypertension.Methods:A study consisted of 22 patients with excessive internal heat syndrome of hypertension, 20 patients with Yin-deficiency Yang excessive syndrome of hypertension was performed. The male patients who have not taken antihypertensive agents or who have taken the agents, but have been on drug withdrawal for more than one week were picked to observe the vital sign and interrelated biochemical indicator of each group. Venous blood samples were collected from patients after fasted for 12 hours. Preloaded for 2 h, the serum samples were obtained by high centrifugation, then, stored in the refrigerator at minus 80℃. GC/TOF-MS was applied to perform PCA and get the metabolic fingerprinting spectra. Multiple statistical analyses were carried out following pretreatment and calculation the integral of the fingerprinting spectra. Scores plot were committed to determine differences between two groups. Meanwhile, principal component analysis (PCA) was used to calculate a basic model for overviewing the data. Partial least squares projection to latent structures & discriminates analysis (PLS-DA) was used to calculate models differentiating groups. Statistical analysis was performed using the SPSS 11.5 software.Results:(1) It is found that there are no significant differences in general states of health, the routine blood tests and the blood biochemical assays between two groups. There are no patent differences on PCA shown in scores plot of two groups, that is to say, principal component of metabolic products from two groups have no remarkable differences. It is demonstrated that there is no obvious differences in Load diagram, but certain differences between two groups were shown in three dimensional image of load diagram. Differentiation compounds can be identified included citrate, oxalate,4,6-dioxoheptanoic acid, glycerate, lactate, ethanolamine, cyclohexanhexol, oleic acid, octadecanoic acid, aspartic acid, threonic acid, and palmitinic acid.Conclusion:It is suggested that metabolomics approach is successful for differentiating excessive internal heat of liver syndrome of hypertension and Yin-deficiency excessive Yang syndrome of hypertension. Although the concrete multivariate regression model can not be completely established between biomarkers of two groups, there is significant differences between one another. It is hopeful that Metabonomics will initially provide objective evidence for TCM categorical identification on hypertension.
