BackgroundsInflammatory response plays an important role in the occurrence and development in the essential hypertension(EH), and can lead to vascular damage and EH target organ damage, how to control high blood pressure from the inflammation point become a new directions in the clinical research. Study confirmed that high blood pressure has relationship with C-reactive protein and tumor necrosis factor-a, sustained high levels of inflammatory mediators C-reactive protein and tumor necrosis factor-a may be the basis for the pathophysiology of EH. JuTeng Capsule is based on traditional Chinese medicine theory and the "different diseases with the same treatment", "the same disease with different treatment" principle, with calming liver yang, Blood and Water-oriented thinking, combined with years of clinical practice, we prepared pure Chinese herbal compound preparation.Clinical observation confirmed that Chinese herbal compound JuTeng Capsule can significantly reduce blood pressure, with significant improvement. To further clarify the clinical effect of JuTeng Capsule mechanism, we used ELISA, immunohistochemistry and other research methods to explore the inflammatory mechanisms of SHR under the treatment of the drug, to provide the experimental basis for clinical application preparation.Research Objective and MethodsObjective: To study the tumor necrosis factor a (TNF-α), C-reactive protein (CRP) expression in heart, brain and kidney tissue and serum content of TNF-αand CRP, to investigate the inflammatory mechanism of anti-hypertension medicine JuTeng Capsule.Methods: Spontaneously hypertensive rats (SHR) 30 were divided into 3 groups of 10 each randomly, were the blank group, JuTeng Capsule group and captopril group. Blank control group were given distilled water, JuTeng Capsule group and captopril group were gavaged with JuTeng and captopril dilution. Blood pressure was measured with a rat tail arterial pressure measurement; using enzyme-linked immunosorbent assay (ELISA) method to detect serum CRP and TNF-αlevels, immunohistochemistry (IH) were used to detect the CRP and TNF-αexpression of heart, brain, kidney tissue. Western blot (WB) to detect the rat brain expression of CRP and TNF-αintensity.Results1. The blood pressure of SHR groups before drug intervention was no difference, 4wk after, the control group increased significantly, while the JuTeng Capsule group and captopril group inhibited the increase of blood pressure (P <0.05).2. The serum ELISA showed that, CRP: JuTeng Capsule (27.8±12.0) ng / L compared with the captopril group (48.7±12.9) ng / L was decreased (P <0.05), 2 groups compared with the control group (71.3±16.4) ng / L were significantly decreased (P <0.05); TNF-α:JuTeng Capsule group (22.5±8.3) ng / L compared with the captopril group (24.0±9.2) ng / L was no significant difference, 2 groups compared with the control group (39.3±16.7) ng / L were significantly decreased (P <0.05).3. Heart tissue immunohistochemistry showed the positive expression specificity is not strong due to the short experimental period, but observations point out that myocardial cells were found in brown clouds in the control group, deeply stained; captopril group smaller and lighter staining; JuTeng Capsule group part of the scattered positive staining, light staining. JuTeng Capsule group, captopril group compared with the control group decreased expression of TNF-αand CRP.4. Kidney tissue immunohistochemical results showed that the control group have the quality of the glomerular membrane stained sheet region, while the drug groups showed superficial punctate staining region, indicating the expression of TNF-αand CRP in the JuTeng Capsule group and the captopril group was decreased.5. Brain tissue immunofluorescence showed that the control group of TNF-αand CRP in the brain cortex region appearing high-density expression, and JuTeng capsule group and captopril group decreased expression.With its semiquantitative western blotting analysis showed that the drug group of brain tissue reduced the expression of TNF-αand CRP intensity. Conclusions1. JuTeng Capsule can effectively combat the rise of spontaneous blood pressure;2. JuTeng Capsule can reduce serum CRP and TNF-αlevels, indicating the function of anti-inflammatory and antihypertension are associated;3. JuTeng Capsule can reduce the expression of CRP and TNF-αin heart, brain, kidney tissue, indicating the target organ protective effects while antihypertension, from this point of view we can explain the inflammatory pathophysiology of JuTeng capsule.4. Inhibition of serum CRP and TNF-αlevels, the improvement of inflammatory state, is JuTeng Capsule pathophysiological mechanisms.
