Objective: To observe the effect of Wei Ao Xin on tumor necrosis factor-α(TNF-α) and C-reactive protein(CRP) of experiment atherosclerosis rats in the serum and artery, and approach that whether it has the effect of anti-inflammatory on the experiment atherosclerosis rats, moreover,to provide the further cognition of its mechanism in the anti-atherosclerosis.Methods: sixty male SD rats were assigned randomly into five groups: group of sham operation(GSO,n=12),group of model(GM,n=12), group of Aspirin(GA,n=12) , group Wei Ao Xin of small dosage (WAXs,n=12), and group Wei Ao Xin of large dosage(WAXl,n=12).After 7 days,the balloon-injured carotid artery angioplasty was performed in the rats of all group except GSO. Then the normal chow was given orally to the rats of GSO. The high fat diet was given orally to the rats of the others. Meanwhile the rats of WAXl were given orally to Wei Ao Xin 200mg/kg.d-1. The rats of WAXs were given orally to Wei Ao Xin 50mg/kg.d-1. Aspirin 50mg/kg.d-1 was given orally to the rat of GA. The drug consumptions were adjusted according the body weight of the rats every 15 days. After three months, serum triglyceride(TG), cholesterol (TC), low density lipoprotein-cholesterol (LDL-C), high density lipoprotein- cholesterol (HDL-C), TNF-αand CRP were measured in all rats. The rats' carotid arteries were stained with hematoxylin-eosin and immunohistochemistry. Image analysis system was applied to measure the area of lumina and the thickness of wall to evaluate the ratio of W to L, and the IOD of TNF-αand CRP . The expression of TNF-αand CRP in the aorta were also measured with Western blot.Results: (1) Compared with group of model ,the level of serum TG,TC and LDL-C in the large dose Wei Ao Xin-treated group were significantly decreased(P<0.05),and lower than the aspirin-treated group(P<0.05). The level of serum TG,TC and LDL-C were no significant difference between the small dose Wei Ao Xin-treated group and the model group(P>0.05). The level of serum HDL-C was no significant difference between the treated groups and the model group. (2) Compared with the model group, the level of serum TNF-αand CRP in the large dose Wei Ao Xin-treated group were significantly decreased(P<0.01),and lower than the aspirin-treated group(P<0.05). The level of serum TNF-αand CRP in the small dose Wei Ao Xin-treated group were significantly decreased(P<0.05).But there were no significant difference between the small dose Wei Ao Xin-treated group and the aspirin-treated group(P>0.05). (3) W/L ratio of carotid artery in the control side had no significant difference among five groups. In the injured side, W/L ratio of carotid artery in the model group was thickened obviously than the sham operation group (P<0.01)and locally formed plaque. The W/L ratio of carotid artery in the large dose Wei Ao Xin-treated group was much thinner than that of model group(P<0.01)and aspirin-treated group(P<0.05). The W/L ratio of carotid artery in the small dose Wei Ao Xin-treated group was much thinner than that of model group(P<0.05). But there were no significant difference between the small dose Wei Ao Xin-treated group and the aspirin-treated group(P>0.05). (4) Immunohistochemistry staining show: Compared with the model group, the IOD of TNF-αand CRP in the large dose Wei Ao Xin-treated group were significantly decreased(P<0.01),and lower than the aspirin-treated group(P<0.05). The IOD of serum TNF-αand CRP in the small dose Wei Ao Xin-treated group were significantly decreased(P<0.05).But there were no significant difference between the small dose Wei Ao Xin-treated group and the aspirin-treated group(P>0.05).(5)The result of Western blot show: Compared with the model group, the level of TNF-αand CRP in the large dose Wei Ao Xin-treated group were significantly decreased(P<0.01),and lower than the aspirin-treated group(P<0.05). The level of serum TNF-αand CRP in the small dose Wei Ao Xin-treated group were significantly decreased(P<0.05).But there were no significant difference between the small dose Wei Ao Xin-treated group and the aspirin-treated group(P>0.05).Conclusion: (1)Wei Ao Xin can reduce the level of serum TG , TC and LDL-C of the experiment atherosclerosis rats. (2)Wei Ao Xin can ameliorate the experiment atherosclerosis rats' carotid artery. Thus it can prevent the development of atherosclerosis. (3)Wei Ao Xin can reduce the levels of serum and arteries TNF-αand CRP in the experiment atherosclerosis rats. (4) Wei Ao Xin can reduce the levels of TNF-αand CRP in the experiment atherosclerosis rat to take the effect of anti-inflammatory.
