| Objective: To observe the effects of Dachengqi Decoction on rats with severe acute pancreatitis.Methods: Fifty male Wistar rats were randomly divided into five groups: sham operation group (A group), SAP group (B group), SAP+ulinastatin group (C group), SAP+Dachengqi Decoction group(D group) and SAP +Dachengqi Decoction+ ulinastatin group (E group),there were ten rats in each group. SAP animal model was induced by retrograde injection of 5% sodium taurocholate, the three treated groups rats were given Ulinastatin in 30min and/or given Decoction in 12 hours followed SAP model set up. The samples of blood, pancreas, lung and ileum tissues were collected at the points of 24 hours after the SAP models made, the pathological changes of pancreas, lung and ileum tissues were observed by microscope with Hematoxylin and Eosin staining. The level of proinflammatory factors such as interleukin-6 (IL-6) and endotoxin (ET) and anti- inflammation factors such as interleukin-10 (IL-10) in the lung, ileum tissues and blood plasma were detected by Enzyme linked immunosorbent assay. Results: The typical pathological changes such as inflammatory cell infiltrate, red blood cell exudation and cellular edema or cellular necrosis were detected in pancreas, lung and ileum tissues of rats in SAP group, and the pathological changes were improve in the treated groups of C, D and E, among them the pathological changes improved mostly in the group E.The level of IL-6, IL-10 and ET in blood plasma,lung and ileum in group B was obviously increased than that in group A(P<0.05).In contrast, the level of IL-10 was increased in groups of C,D,E comparing to that in group B(P<0.05),and it is increased mostly in group E, and there is no difference between in C and D groups(P>0.05).Conclusion: Dachengqi decoction could affect inflammatory reaction of severe acute pancreatitis by reducing proinflammatory factors such as ET and IL-6,and increase anti-inflammatory factor as IL-10,Dachengqi Decoction and ulinastatin have additive or synergetic effects on anti-inflammatory action in rats with severe acute pancreatitis. |
