Objective: To detect the differential expression of ZIP in benign prostate hyperplastic and prostate cancer, investigate the possible correlation between the expression of ZIP in prostate cancer ,prostate-specific antigen (PSA) and t?he tumor's Gleason score , evaluate the clinical significance of ZIP for the diagnosis of prostate cancer.Materials and Methods: All paraffin-embedded sections in our research were collected from the patients who treated with TUEVP (transurethral electrovaporzation of prostate) or radical prostatectomy in our hospital from 2007-2009, in which there were 15 cases of benign prostate hyperplasia and 45 cases of prostate cancer. Use light microscope to observe the pathological changes of prostate cells by HE staining. Immunohistochemistry to detect the expression of ZIP1,ZIP2,ZIP3 in BPH and PCa, and to investigate the possible correlation between the expression of ZIP in prostate cancer ,prostate-specific antigen (PSA) andt?he tumor's Gleason score. All data were processed by SPSS version 17.0 analysis software.Correlation between PSA and ZIP analyzed by Spearman analysis program to calculate the correlation coefficient r value.Results: According to pathological results, the patients were divided into benign prostatic hyperplasia and prostate cancer group. Immunohistochemistry detection of the expression of ZIP in the two groups: the positive rate of ZIP1, ZIP2, ZIP3 in BPH was 73.33%, 93.33%, 80.0%, respectively; but in PCa , the positive rate was 8.89%, 11.11%, 8.89%, Line×listχ2 test or Fisher exact test statistical analysis,χ2= 21.60, 35.148, 29.091, P less than 0.05, ZIP1, ZIP2, ZIP3 expression in the two groups statistically significant difference. In this study, ZIP1, ZIP2, ZIP3 in prostate cancer patients with low level of expression. The relationship between ZIP protein and serum PSA: positive expression of ZIP in the PSA <10ng/ml group was significantly higher than in PSA > 10ng/ml group, P less than 0.05, statistically significant is difference. There is a negative correlation between ZIP and PSA(r=-0.367,.-0475,-0.376).According to pathological Gleason score of prostate cancer, the positive rate of ZIP proteins in the three groups, Gleason score group 2 to 4, 5 to 7 and 8 to 10,has no significant difference. Between the three groups were statistically analyzed, P more than 0.05,that ZIP protein and prostate cancer pathological Gleason score had no significant difference, indicating that ZIP proteins in prostate cancer early began to reduce or even missing. In this experiment, as the diagnostic indicator for prostate cancer, using combined PSA greater than10ng/ml with negative expression of any ZIP, compared with a single PSA greater than 10 ng / ml or negative expression of any ZIP, has a higher specificity (93.33%). According to the list×lineχ2 test, P less than 0.05, and the specificity of three diagnostic indicators had significant statistical difference.Conclusions:1. The positive expression of ZIP protein in prostate cancer was significantly lower than that of prostate.2. The relationship between ZIP protein and serum PSA: positive expression of ZIP in the PSA <10ng/ml group was significantly higher than in PSA > 10ng/ml group.There is a negative correlation between ZIP and PSA.3. ZIP protein and prostate cancer pathological Gleason score had no significant difference.4. Combined PSA greater than10ng/ml with negative expression of any ZIP, as the diagnostic indicator for prostate cancer, can increase the diagnostic specificity.
