The flowers and buds of Lonicera japonica Thunb., commonly known as 'erhua' in Chinese traditional medicines, are used for their latent-heat-clearing, antipyretic, detoxicating and antiinfammatory actions to treat affection by exopathogenic wind-heat or epidemic febrile diseases at the early stage, sores, carbuncles, furuncles and swellings. The modern pharmacal researches show that it can antagonize pathogenetic microorganism, anticatarrhals, decline fever, hemostasis, protect liver, adjust immune function, lower blood-lipid , stimulate central nerves, antioxidation, anticancer and so on. In clinic, it is used for curing some sorts of infections, many kinds of catarrhals, high fever, high blood-lipid and arresting dysentery. On the other hand , studies also indicate its moderate anti-HTV activity.In this paper the water-extracts of Lonicera japonica Thunb.(from Fengqiu,He-nan) were chromatographed over macroporous pore resin column, Sephadex LH-20, ODS, silica gel column, then 16 compounds were separated and purified. On the basis of their spectral data (UV, IR, NMR, MS etc.) and chemical evidence, their structures were identified. These compounds contain 9 secoiridoids, 5 organanic-acids, 2 long chain adipose alcohols and 1 aldehyde. We did proper configurable embellish to mass compounds and lay a foundation for the relation of structure-activity. Most of these compounds are also evaluated in biological activities such as anti-tumour activity, immune activity and so on.Firstly, researches on the chemical constituents of Lonicera japonica were reviewed from the aspects of their species-resource, chemical components, technology of extraction and isolation, pharmacological activities, quality standards, application and development.Secondly, we studied systemically on Lonicera japonice extraction by water. 16 compounds were isolated by using various means and were elucidated by their spectral data.The confirmed compounds were Secologanic acid (1) , Vogeloside(2), 7-epi-vogeloside(3), 7-hydroxy-methyl-vogeloside(4), Sweroside(5), Secologanoside(6), Secoxyloganin(7), (E)-aldosecologanin(8), 2(E)-3-ethoxy acrylic acid(9), Protocatechuic acid(10), Caffeicacid(11), Ferulic acid(12), Chlorogenic acid(13), Pentacosa alcohol(14), Pentatriaconta alcohol(15), 2-(2-propenyloxy)-ethanal(16). Among them, compound 1, 4, 6, 9, 10, 12, 14, 15, 16 were separated for the first time from Lonicera Japonica Thunb. In addition compound 1 was acetylated, then a kind of all acetylated derivative was obtained. It was important for the study on the relation of structure-activity.Thirdly, the research progress on natural iridoid compounds were summarized because of a mount of secoiridoids isolated from Lonicera japonica. We hope that it will be best used in clinic practice and new drug development.
