The Application Of CTB-HRP In The Research Of The Properties Of Relationship Between Acupoints And Between Acupoints And The Viscera-related Nerve

Objective:To reveal the acupoints and the specific relation between the acupoints and attribute of the nerve related to viscera with cholera toxin subunit B conjugated with Alexa Fluor 488, 594(CTB-Alexa 488, 594) tracer technique, to reveal the structural features and relation in three levels of single acupoints, acupoints to acupoints and acupoints to viscera.Method:18 adult rats weighted 200-280g, fed in a 12hours light and 12 hours dark environment, given free access to food and water, randomly divided in to group 1, group 2 and group 3, were micro-injected with CTB-Alexa 488 or CTB-Alexa 594. Group 1 were micro-injected in single acupoints. Group 2 were micro-injected in 2 acupoints. Group 3 were micro-injected in single acupoint and related viscera. And 3 groups were perfusion fixed and drew materials from posterior root ganglion, spinal cord and brain to observe after 72 hours'feed.Result: Experiments confirmed that the segmental and regional distribution of the spinal column and the nutrition characteristic of acupoints is clear. The 46 single labeled neuron in lumber ganglion mainly concentrated in L4/L5, and the neuron in the lumber spinal anterior horn is mainly concentrated in L5; 330 double labeledneuron in cervical spinal in mainly concentrated in C7/C6, of which the neuron in anterior horn is mainly concentrated in C8. We only found single labeled neuron inDRG in acupoints and viscera labeling, it is the same case in spinal marrow. It also proves that fluorescent tracer technique is simple, quick and easy to manipulate. Conclusion:The segmental and regional distribution trait of the acupoints in body surface and the therapeutic effect of acupoints have relation to nerve system. The two acupoints marking experiment prove not only the nervous segmental and regional trait of acupoints but also the acupoint to acupoint relation in nervous anatomy probably due to that the same neuron govern two different body surface region. The result provides anatomical proof to the acupuncture therapy combined with yuan point and luo point.
