The theory of meridian viscera correlation is an important part of the theory of meridians and collaterals.It is a systematic theoretical summary of the relationship between the body surface and viscera and the mechanism of interaction between meridian theory and viscera,which can directly and effectively guide the clinical diagnosis and treatment.In recent years,it has become a hot research topic,and has been funded by major national scientific research projects.The previous studies on the relationship between acupoints and viscera have been mainly focused on the positive argumentation from acupoints to viscera,that is,stimulating the acupoints on the body surface,detecting the relative visceral hemodynamics,electrophysiology,vasoactive substances and molecular biological indexes.And from the peripheral and central nervous system,activation of cerebral cortex division,protein gene and other levels of action from outside to inside to study the relationship between the two,but by pointing to the surface point by the organ,the research on reverse demonstration inside is relatively weak,mostly concentrated in the one body state,the relevant acupoints,non related acupoints difference in the morphology,sound,electricity,light,hemodynamics and molecular biology etc.But when the viscera is in different states,there are few studies on the specific changes of the related and uncorrelated acupoints.At present,most of the research models of the meridian viscera correlation are focused on heart diseases,especially myocardial ischemia model is the most mature,and some experimental studies have proved that low-frequency electroacupuncture can obviously improve myocardial ischemia in perfusion injury,and can regulate vasoactive substances.And explain its mechanism of action from the level of sympathetic nerve.It is suggested that electroacupuncture can improve cardiac function by inhibiting the excitability of cardiac sympathetic nerve and enhancing the excitability of A ? and C afferent nerves.In this study,the myocardial ischemic injury model of rats was used to record the changes of body surface temperature in related acupoints Neiguan and non-related acupoints Zusanli,Yanglingquan when in the injured state and the low frequency electroacupuncture acupoints,non acupoint treatment state,The influence of frequency of low frequency electroacupuncture intervention on it was also studied.The content of nitric oxide(NO)/calmodulin(CaM),a sympathetic nerve related molecule in the tissue of acupoint area,was detected in order to reveal the difference of temperature effect of low frequency electroacupuncture on related acupoints and non-related meridians?the law of the response of the relevant acupoints to the changes of the state of the viscera.This paper tries to reveal the intrinsic mechanism of temperature specific change of meridian from the molecular mechanism of sympathetic nerve and enrich the scientific connotation of the theory of meridian-viscera correlation.Objective:(1)To compare the effects of low frequency electroacupuncture intervention on the temperature of the related acupoints and uncorrelated acupoint in rats with myocardial ischemia,and to find out the most sensitive and specific acupoint point to responses to different myocardial states(normal physiological state,ischemic injury state,low frequency electroacupuncture at acupoint state,low frequency electroacupuncture at non-acupoint state)(2)Selecting the acupoints of the relevant and uncorrelated acupoint which are the most and the least able to respond to the temperature specific changes in the rat myocardium in different states.The contents of NO and CaM were detected by ELISA method and nitrate reductase method in order to explore the intrinsic mechanism of the temperature change of acupoint when the state of viscera was changed.To provide experimental basis for the relationship between acupoints and viscera.Methods:80 male Wistar rats were divided into two groups according to random number table:one day and three days group,then divided into 5 groups according to random number table,8 in the normal group,8 in the sham operation group,the remaining 24 in the myocardial ischemia model rats were equally distributed in the model group,Low frequency electroacupuncture acupoint group and low frequency electroacupuncture non-point group,8 rats in each group.After the first and third times of electroacupuncture treatment,the skin temperature of every group of rats was detected by infrared thermal imaging technique,and the skin temperature of each group was analyzed statistically.According to the statistical changes conditions,the points which are the most effective and the most unresponsive to the changes of heart specificity were selected,and the content of NO and CaM in the tissues of the acupoints were detected by ELISA and nitrate reductase methods.Results:(1)Compared with the control group,Treatment once and three times,the electrocardiogram"J" point difference in the model group was significantly higher than that in the control group(P<0.01,P<0.01).Compared with the model group,and the "J" point difference in the low-frequency electroacupuncture at acupoint group was significantly lower than that in the model group(P<0.01,P<0.01).(2)Longitudinal comparison of the temperature changes in the non-related acupoints of Zusanli and Yanglingquan and the related acupoints Neiguan when in different states of myocardium:Compared with the blank group,when myocardial injury was observed in the model group,the related acupoints of "Neiguan" in the 1st and 3rd groups decreased significantly(P<0.01,P<0.0l),the temperature of non-related acupoint "Zusanli" in the 3-day group decreased(P<0.05).Compared with the model group,the temperature of the "Neiguan"point increased on the 1st and 3rd day when the low frequency electroacupuncture meridian was in the state of intervention(P<0.01,P<0.01),but the temperature of each point area did not change significantly in the non-point intervention state of low frequency electroacupuncture group.The difference of changes between the groups under different times of electroacupuncture intervention was compared.The results showed that there was no significant difference between the one and three times of acupuncture and moxibustion.(3)Compared with the control group,for treatment once and three times,the contents of NO at"Neiguan" point in the model group were all decreased(P<0.05,P<0.05)and the contents of CaM were all decreased too(P<0.05,P<0.05).Compared with the model group,after the intervention of low frequency electroacupuncture at acupoint,the content of NO in "Neiguan"acupoint was all increased(P<0.05,P<0.05).and the content of CaM was also increased(P<0.05,P<0.05),but there was no significant change in NO and CaM in the acupoint area of Yanglingquan.There was no significant difference in NO/CaM content between 1 and 3 times.Conclusion:(1)The low frequency electroacupuncture Neiguan can improve the temperature drop on the related acupoint Neiguan of myocardial ischemia rats,and have no statistical significance on the change of the temperature of the non-related acupoints.(2)Low frequency electroacupuncture at Neiguan can increase the content of NO and CaM in the related acupoint Neiguan,Associated with the sympathetic response which can suggesting may be the mechanism of temperature changes in related acupoint Neiguan is related with the state of the viscera.(3)1 times and 3 times of low frequency electroacupuncture had no significant difference in the improvement of myocardial condition and the response of related and unrelated acupoints.(4)There were specific changes in the temperature and NO/CaM content of the related acupoint Neiguan Points in different myocardial States,in the three Acupoints,The most obvious changes is in related acupoint Neiguan,Zusanli second,which indicated that the related acupoint were more specific than non related acupoints to respond to the changes of the related viscera.
