Abstract/Summary:
Objective:To study the effect of chronic intermittent hypoxia on the behavior and the oxidative stress in corpus striatum of parkinson's disease model mouse. Methods: Forty-four six-week-old C57BL/6 male mice were randomly divided into four groups: paraquat (PQ) group, chronic intermittent hypoxia (CIH) group, PQ+CIH group and control group. The level of dopamine(DA) and the activity of superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX), the level of glutathione and malondialdehyde (MDA) in corpus striatum.were examined. Results:At the beginning of the experiment, the scores of pole test in each group the difference had not statistical significance; At the end of the experiment, compared with control group and at before of the experiment of PQ group, CIH group and PQ+CIH group respectively, the scores of pole test were decreased; Compared with PQ group, the scores of pole test was decreased in PQ+CIH group; At the beginning of the experiment, the difference in the spontaneous motor activity in the open field in each group did not have statistical significance; At the end of the experiment, compared with control group and at before of the experiment of CIH group, PQ group and PQ+CIH group respectively, the ambulation and the rearing were decreased; Compared with PQ group, the ambulation and the rearing were significantly decreased in PQ+CIH group.Compareing the level of DA in the striatum:the control group was significantly higher than the PQ group and PQ+CIH group,but was not significantly higher than the CIH group. Compared with the PQ group, The PQ+CIH group was significantly reduced; Compareing the activity of SOD in the striatum:the control group was significantly higher than the PQ group, CIH group and PQ+CIH group (P<0.05) (63.01±2.56,59.35±3.20,60.37±3.37,55.51±2.92) U/ml; The PQ Group Compared with the CIH group and PQ+CIH group, the PQ group and the CIH group was not significantly different,the PQ+ CIH group was significantly reduced; compared with the CIH group, PQ+CIH group also significantly reduced. Compareing the activity of GSH-PX in the striatum:the control group was significantly higher than the PQ group, CIH group and PQ+CIH group (P <0.05)(55.68±5.10,50.07±5.61,51.95±3.95,46.20±2.59) U/gprot; The PQ Group compared with the CIH group and PQ+CIH group, the PQ group and the CIH group was not significantly different, the PQ+CIH group was significantly reduced; compared with the CIH group, the PQ+CIH group also significantly reduced. Compareing the level of GSH in the striatum:the control group was significantly higher than the PQ group, CIH group and PQ+CIH group (P<0.05)()(16.60±1.62,14.50±1.25,13.82±1.08,12.52±0.73) mg/gprot; The PQ Group compared with the CIH group and PQ+CIH group, the PQ group and the CIH group was not significantly different, the PQ+CIH group was significantly reduced; compared with the CIH group, PQ+CIH group also significantly reduced. Compareing the level of GSH in the striatum:the control group was significantly lower than the PQ group, CIH group and PQ+CIH group (P<0.05) (2.97±0.38,3.41±0.58,3.90±0.83,4.57±0.86) nmol/mgprot; The PQ Group compared with the CIH group was not significantly different, the PQ Group compared with The PQ+CIH group was significantly reduced; the CIH group compared with PQ+CIH group also significantly reduced.Conclusion:CIH increased the damage of behavior and reduced the level of DA in striatal of PD mice, which may be associated with increased oxidative stress in striatum of PD model mice.
