Objective: In China, renal cell carcinoma is one of the severe disease that threatens people health,is the mostest of the Urological tumor. Recently years, ADM in ovarian cancer, lung cancer and other tumors of the occurrence, development has been a growing concern, its in renal cell carcinoma effect also has begun by widespread interest and research. Adrenomedullin (ADM) is the first discoverd from Japan scholars in 1993 from the pheochromocytoma in separating out of a peptide vasodilator, its consists of 52 amino acid composition, it and calcitonin gene-related peptide (CGRP Calcitonin-gene related peptide,) homology between 24%,it have typical disulfide bridge, belonging to the family of CGRP. Current clinical studies show that ADM in many cancer patients in overexpression, and histologic grade and malignant grade interrelatedtothe, and with the poor prognosis. Adm in the body can play many complex biological effects. Adm activate adenosine guanylyl Cyclase- PKA signal transduction pathway, so that the cells in cAMP levels rise; Adm improves intracellular Ca2+concentration-induced vasodilation and activated endothelial nitric oxide (NO) synthase; Adm activated MAPK signaling to the paths to promote cell mitosis, etc. These are likely to occur with the tumor development is closely.Hypoxia is solid tumor growth have common features, low oxygen stimulate tumor cells produce some columns of the reaction to promote tumor growth and progress. Studies show that low oxygen can stimulate the ADM in the expression, subsequently many reports in low oxygen environments in a variety of tumor cells in ADM overexpression. Low-oxygen environment ADM overexpression and its biological effects on tumor growth development process plays an important role.This study in human renal cell carcinoma as object, given the relative low oxygen stimulate processed observing its expression level of ADM, further in renal cell carcinoma validation hypoxia factors on the influence of ADM. However, little attention has been paid to the ADM in renal cell carcinoma, as a result of this research to validate and exploring the possibility of renal cell carcinoma development mechanism and the clinical diagnosis of renal cell carcinoma and reasonable treatment provides theoretical basis.Methods:Human Renal carcinoma cell 786-O Peking Union Medical CollegeCulture of Renal carcinoma cell line and give different conditions of the respectively low-oxygen environment,use of proteins imprinted (Western blot) detection of various groups of Renal carcinoma cell line expression levels in ADM.ADM protein expression levels with independent sample t-test method or test for variance analysis. Statistical significances were determined at level of P<0.05. The statistical package was SPSS13.0.Results:1 Various conditions of low oxygen stimulate protein expression levels in ADMβactin protein: The molecular weight is 42kD. After Western hybridi- zation, 42kD bands were found.βactin protein was detected when ADM protein were examined and as the standard for semiquantitative analysis of ADM protein expression.ADM protein: The molecular weight is 7kD. After Western hybridization, 7kD bands were found. The bands were analyzed by gel image analyzing system. The protein expression was semiquantitative data (named IOD), and the relative IOD was used for the comparison of protein expression. Comparing to normal tissues, various conditions of low oxygen stimulate group cell lines have different degrees of ADM high expression, and correlation with stimulating time experiencing.Conclusion:Low-oxygen environment, can contribute to renal cell carcinoma in the expression of ADM, and with low oxygen stimulate time experiencing correlations. But low-oxygen environment to promote the expression of specific ADM mechanism and ADM in renal cell carcinoma and specific biological effect is still the need for further in-depth discussion of.
