| Objective:To discuss the influence of pre-injected XiangDan Parenteral Solution into PC6 on endothelial injury and endothelial secretion by testing the coronary arteries'Bcl-2 (B cell lymphoma/lewkmia-2),Bax (Bcl-2 associatied X protein),Bcl-2 and eNOS(endothelial nitric oxide synthase) on the rat model. What's more, it maybe helpful to provide some new ideas and methods for clinical treatment of coronary disease and myocardial ischemia.Methods:SD rats were given Xiangdan parenteral solution into PC6 for 14 days before myocardial ischemia model was induced by isoprenaline injected celiac-subcutaneously. Then the myocardial tissue and coronary artery were observed for pathological changes; the Bax,Bcl-2,eNOS of coronary arterial endothelium were also detected with immunohistochemical method.Result:(1) The typical pathological changes of myocardial ischemia and hypoxia in the model group were observed. The acupuncture combined with medicine group can significantly protect the myocardial tissue from ischemia and hypoxia, narrow the area of cardiac damage, reduce the degree of myocardial ischemia, and speed up restoration. The acupoint injection group and drug control group have little effect on protecting rats from myocardial ischemia. (2)The thickened-wall and narrowed-cavity of coronary artery were observed in the acute myocardial ischemia rats. The acupuncture combined with medicine group, acupoint injection group and drug control group can alleviate the thickening degree of coronary artery wall, and there's no difference among them. (3) The content of Bax increased after myocardial ischemia. However, all of the three groups could decrease it, within which acupuncture combined medicine group functioned most significantly. (4) The expression of eNos decreased after myocardial ischemia. However, all of the three groups could increase it, within which acupuncture combined medicine group functioned most significantly. (5) The expression of Bcl-2 decreased after myocardial ischemia. However, all of the three groups could increase it, within which acupuncture combined medicine group functioned most significantly.Conclusions:The method of pre-injected XiangDan Parenteral Solution into PC6 can protect the myocardium from ischemia and hypoxia. It can also alleviate the thickening degree of coronary artery wall, reduce Bax level, and increase Bcl-2 and eNos content, it may be lower the damage of myocardial ischemia by improving endothelial secretion and inhibiting endothelial cell apoptosis.
