Study On The Effect And Mechanism Of Liushen Pill On The Antitumor Action In Nude Mice

Objective:To investigate the inhibitional effect of Liushen Pill on the human Esophagus squamous cell carcinoma xenografts of the nude miceMethod:42 nude mice were randomly divided into 6 groups:high dosage of Liushen Pill group,middle dosage group,low dosage group,DDP group,model group,blank group,then established the human Esophagus squamous cell carcinoma xenografts model which were treated by dosage regimen,dynamic observed the tumor growth in different groups,record living conditions,measured the tumor volume,draw growth curve, the nude mice were sacrificed after 20 days treatment,Neoplasm weight was taken and the tumor-suppressing rates was calculated; Immunohistochemistry was used to detect microvessel density(MVD),the expression of apoptosis index(AI), VEGF and Bcl-2.Result:Liushen Pill can inhibit the growth of the human Esophagus squamous cell carcinoma xenografts.The weight of tumor in middle dosage group,low dosage group and DDP group was much lower than that of the model group(P<0.01), compared with model group, the difference between model and high dosage group was insignificant (P>0.05), The microvessel density(MVD) in the treatment group was much lower than that of the model group, meanwhile the apoptosis index(AI) was much higher than that of the model group, The high,middle,low dosage group can effectively down-regulate the expression of VEGF,Bcl-2.Conclusion:1 Liushen Pill can inhibit the growth of the human Esophagus squamous cell carcinoma xenografts.2 Liushen Pill treatment can effectively down-regulate the expression of VEGF and MVD,then inhibit the angiopoiesis,which is one of the tumor-inhibiting mechanism of Liushen Pill.3 Liushen Pill can inhibit tumor by down-regulating the expression of Bcl-2 and promoting apoptosis.
