| Objective To investigate the expression of NGF and TrkA in the ectopic and eutopic endometrium of women with adenomyosis and their relationships with dysmenorrhea.Methods Ectopic endometrium and eutopic endometrium from 30 patients with adenomyosis (Group adenomyosis) and endometrium from 19 patients with CIN (â…¡-â…¢) without dysmenorrhea Group control were collected. It was performed preoperative that the dysmenorrhea degree questionnaire for the patients with adenomyosis (Group mild dysmenorrhea 15 cases and Group severe dysmenorrhea 15 cases) Immunohistochemistry and RT-PCR were used to detect the protein and mRNA of NGF and TrkA of the all the specimens respectively, and to examine the correlations between the expression levels of NGF and TrkA and the pain intensity.Results For the expression levels of NGF and TrkA of ectopic and eutopic endometrium, the Group adenomyosis were higher than the Group control (P=0.000).No difference of the expression levels of NGF and TrkA was found between the ectopic and eutopic endometrium of Group adenomyosis (P=0.293; P=0.919).There were positive correlations between the expression levels of NGF of the ectopic and eutopic endometrium of Group adenomyosis and the pain intensity (r =0.637 P=0.000 r = 0.487 P=0.006). The NGF expression level of the ectopic endometrium of Group severe dysmenorrhea was higher than those of Group mild dysmenorrhea (P=0.001) and Group control (P=0.000). The NGF expression levels of the ectopic endometrium of Group mild dysmenorrhea was higher than those of Group control (P=0.039). The NGF expression level of the eutopic endometrium of Group severe dysmenorrhea was higher than those of Group mild dysmenorrhea (P=0.003) and Group control (P=0.000). The NGF expression level of the eutopic endometrium of Group mild dysmenorrhea was higher than those of Group control (P=0.013).There were positive correlations between the TrkA expression level of the ectopic endometrium of Group adenomyosis and the pain intensity (r = 0.662 P=0.000). The TrkA expression level of the ectopic endometrium of Group severe dysmenorrhea was higher than those of Group mild dysmenorrhea (P=0.000) and of Group control (P=0.000). The TrkA expression level of Group mild dysmenorrhea was higher than those of Group (P=0.045). There were no positive correlations between the TrkA expression level of the eutopic endometrium of Group adenomyosis and the pain intensity (r = 0.352 P=0.057). No difference of TrkA expression level was found between Group severe dysmenorrhea and Group mild dysmenorrheal (P=0.490).Significant difference of TrkA expression level was found between Group severe dysmenorrhea and Group control, and also found between the Group mild dysmenorrhea and Group control (P=0.000).For the levels of TrkA mRNA and NGF mRNA, there were no difference among the ectopic and eutopic endometrium of Group adenomyosis and those of Group control (P = 0.235; P = 0.218). The levels of NGF mRNA and TrkA mRNA of the ectopic endometrium of Group adenomyosis didn't correlated wih dysmenorrhea degree r = -0.242 P=0.197 r = 0.250 P=0.182 ; neither did those of the eutopic endometrium of Group adenomyosis r = 0.341 P=0.065 r = 0.108 P=0.569 . Among Group severe dysmenorrhea, Group mild dysmenorrhea and Group control the expression levels of NGF mRNA and TrkA mRNA in eutopic or ectopic endometrium had no significant correlation(P=0.184, P=0.600, P=0.090, P=0.349, respectively). Conclusion The NGF and TrkA express highly in adenomyosis, and they may play an important role in the pathogenesis and the development of adenomyosis,The expression levels of NGF and TrkA in adenomyosis may be affected after the transcription, thus increasing the protein expression.The dysmenorrheal of adenomyosis positively correlated with the expression NGF and TrkA in the ectopic and eutopic endometrium of adenomyosis. |
