Objective:To observe the influence of the neural and motor function, muscular tone and GAT-1 in brain stem, cervical enlargement as well as intumescentia lumbalis by puncturing on points locating on yin and yang heel vessels. Initially investigate the possible mechanism of body-spasm improvement after stroke by puncturing on points of yin and yang heel vessels..Methods:Methods:60 SD rats with weight from 250-300g were randomized to 4 groups: control(n=15), sham operation (n=15), model (n=15) and electroacupuncture (n=15). Suture Method was administered to model and electroacupuncture groups for establishing MACO mode, while the sham operation group was only given discission of right-side arteria carotis communis, internal and external carotid. Control group wasn't given any interventions.3 days after modeling, the electroacupuncture group was given the electroacupuncture treatment on Shen Mai (BL 62) and Zhao Hai (KI 6) which locate on heel vessels,30 min every day,6 days in total. While the normal,sham operation and model groups were free. Before and after modeling, Zea-Longs standard was adopted to assess neural function impairment and recovery condition at different time. Muscular tone was measured by BL-420E+four-channel physiological recorder and Ashworth evaluation standard. Killed the mice when the treatment was completed and tested tissue of brain, cervical enlargement as well as intumescentia lumbalis with IHCA for each group, to know the express of GAT-1 in those tissues.Results:(1) Zea-Longs standard of neural damage evaluation:After MACO modeling, control and sham operation group no neural damage syndrome, while model and electroacupuncture group with different neural damage level; after treatment, though the score of electroacupunture group lower than model group (P<0.05)(2) The result of muscular tone assessment:First day after modeling, there's no significant difference between sham operation and control group, and in comparison of model and electroacupuncture group there was significant difference in reduction of muscular tone between those two groups and sham operation one; the third days after modeling, there was significant difference in increase of muscular tone between those two group and sham operation one (P<0.01), which demonstrated that the muscular tone of mice experienced from ascending to descending process after receiving MACO surgery.9 days after modeling, especially remarkable in electroacupunture group with difference between model group (P<0.05)(3) IHCA of GAT-1:Brainstem, cervical enlargement and intumescentia lumbalis of each group had positive GAT-1 cell expression. Compared with sham operation and control groups, the number of positive cell in brainstem, cervical enlargement and intumescentia lumbalis had significant growth in model and electroacupuncture groups (P<0.01). post-treatment GAT-1 expression of brainstem, cervical enlargement and intumescentia lumbalis tissues in electroacupuncture group were lower than model group, with remarkable difference (P<0.01)Conclusions(1) Electroacupuncture treatment on acupoints along heel vessels has a promoting effect on neural damage symptom in MACO mode mice; and also can regulate the expression of GAT-1 in brainstem, cervical enlargement and intumescentia lumbalis down. Moreover, it can reduce the muscular tone of spasm body with hemiplegia.(2)One of potential mechanism of promoting body spasm after stroke with electroacupuncture is that inhibit absorption of GABA by down regulating the release of GAT-1 from central nerve to increase GABA concentration in synaptic space for reinforcing the inhibition function.
