Objects: To detect the expression of the MTA1, HIF-1αand MMP-9 in the normal cervical epithelial tissue(NCE), cervical intraepithelial neoplasia (CIN) and squamous cell carcinoma of cervix (SCC) , and examine the possible roles and clinical significance of MTA1, HIF-1αand MMP-9 in the occurrence and progression of cervical cancer.Methods: The expression of the MTA1, HIF-1 and MMP-9 protein in 60 cases of squamous cell carcinoma of cervix (SCC), 30 cases of cervical intraepithelial neoplasia (CIN) and 30 cases of normal cervical epithelial tissue was detected by using immunohistochemistry EliVisionTM plus (two-step) method. The expressions of MTA1, HIF-1αand MMP-9 in cervical cancer and the clinicopathological parameters of cervical cancer were analyzed to examine the possible roles and clinical significance of MTA1, HIF-1αand MMP-9 in the occurrence and progression of cervical cancer..Results: 1.The positive expression rate of MTA1 in NCE,CIN and SCC was 13.33%, 43.33%, 71.67% respectively. The positive expression rate of MTA1 in CIN was significantly higher than that in NCE , the difference was statistically significant (P<0.05). The positive expression rate of MTA1 in SCC was significantly higher than that in CIN and NCE, the differences both were statistically significant (P<0.05). Overexpression of MTA1 in SCC was correlated with lymph node metastasis , the depth of infiltration and vascular involvement. The relationship of MTA1 protein with clinical stage and histopathological grading was not found.2. The expression of HIF-1αwas not found in NCE, and the positive expression rate of HIF-1αin CIN and SCC was 36.67%, 75% respectively. The positive expression rate of HIF-1αin CIN was significantly higher than that in NCE, the difference was statistically significant (P<0.05). The positive expression rate of HIF-1αin SCC was significantly higher than that in CIN and NCE, the differences both were statistically significant (P <0.05). Over-expression of HIF-1αwas correlated with lymph node metastasis, histopathological grading, the depth of infiltration and vascular involvement. The relationship of HIF-1αwith clinical stage was not found.3. The positive expression rate of MMP-9 in NCE,CIN and SCC was 16.67%, 53.33%, 81.67% respectively. The positive expression rate of MMP-9 in CIN was significantly higher than that in NCE, the difference was statistically significant (P <0.05). The positive expression rate of MMP-9 in SCC was significantly higher than that in CIN and NCE, the differences both were statistically significant (P <0.05). Overexpression of MMP-9 in SCC was correlated with histopathological grading, lymph node metastasis, the depth of infiltration and vascular involvement. The relationship of MMP-9 protein with clinical stage was not found.4. The positive expressions between MTA1 and HIF-1α, MTA1 and MMP-9, HIF-1αand MMP-9 were significantly positive correlation in SCC (r = 0.320, 0.371, 0.323, P<0.05).Conclusion: Overexpressions of MTA1, HIF-1αand MMP-9 are found in cervical cancer,and are closely related with malignant biological behaviors of cervical cancer such as invasion and metastasis. Overexpression of MTA1, HIF-1αand MMP-9 may play some important roles in occurrence and development of cervical cancer, all of them may play synergetic roles.
