Objectives To study the expression of squamous cell carcinoma antigen(SCCA),tumor metastasis related factor-1(MTA1),multiple tumor suppressor gene(P16)and nucleusassociated antigen(Ki-67)in cervical lesions.To study the correlation between the four factors and the infection high-risk HPV infection.Methods 123 patient with cervical lesions in the Affiliated Hospital of North China University of Science and Technology during March 2015 to November 2016 were entrolled in the study.the cases including 31 cases of low-grade squamous intraepithelial lesions(LSIL),31 cases of high-grade squamous intraepithelial lesion(HSIL)and 29 cases of cervical cancer.While the control group including 32 cases with chronic cervicitis.All the pathological results were confirmed by pathologist,and none of them has received radiotherapy,chemotherapy,hormonotherapy.Collected the clinical datas and the cervical tissues which were gotten in the process of surgery or colposcopic biopsy,collect the cervical secretions of these patients,using cobas 4800 detection technology to detect DNA in the cervical secretory HPV virus.Immunohistochemistry(S-P method)was used to detect the expression levels of SCCA,MTA1,P16 and Ki-67 in different cervical tissues,the results of immunohistochemistry in two different cervical lesions were compared by rank sum test.Western blot was used to detect the expression of SCCA and MTA1 in different cervical lesions,the results of Western blot analysis between two different tissues were analyzed by independent sample t test.And the correlation between the four factors and high-risk HPV infection was analyzed by Spearman rank correlation test.Results 1 Immunohistochemistry showed that SCCA protein was strongly positive in cervical squamous cell carcinoma,positive expression in HSIL ? LSIL,and weakly expressed in chronic cervicitis.Pairwise comparison,HSIL and LSIL were higher than those in chronic cervicitis group(P<0.05).The expression level and staining intensity of HSIL group were higher than LSIL,but there was no significant difference between them(P=0.197).The data were assessed by the Spearman correlation coefficients,?=0.640(bilateral test P<0.05).The results of Western blot showed consistent with the results of immunohistochemistry,showing that the expression level and staining range of SCCA protein increased gradually with the increase of cervical lesions.2Immunohistochemical results showed that MTA1 protein in cervical squamous cell carcinoma and HSIL showed positive or strong positive expression,in LSIL and chronic cervicitis group showed weak expression.Pairwise comparison,the expression of MTA1 in cervical squamous cell carcinoma was higher than that of HSIL,LSIL group(P<0.05);The expression level and staining range of HSIL group were higher than those of LSIL,HSIL group was higher than that of LSIL and chronic cervicitis group(P < 0.05);the expression of MTA1 in LSIL group was higher than that in chronic cervicitis group,but there was no significant difference between them(P>0.05).The data were assessed by the Spearman correlation coefficients,?=0.635(bilateral test P < 0.05).Western blot results were consistent with immunohistochemical results,showing that the expression level and staining range of MTA1 protein increased gradually with the increase of cervical lesions.3P16 protein was strongly expressed in cervical squamous cell carcinoma,In HSIL,LSIL positive expression and in the chronic cervicitis group was weakly expressed.Pairwise comparison,he expression level and staining range of P16 in cervical squamous cell carcinoma were higher than those in HSIL and LSIL groups(P<0.05).LSIL and HSIL were higher than those in chronic cervicitis group(P<0.05);The data were assessed by the Spearman correlation coefficients,?=0.784(bilateral test P < 0.05).Showed that the level of P16 protein and the range of staining increased gradually with the increase of cervical lesions.4 Ki-67 protein was strongly expressed in cervical squamous cell carcinoma,In HSIL,LSIL positive expression and in the chronic cervicitis group was weakly expressed.Pairwise comparison,the expression level and staining range of Ki-67 in cervical squamous cell carcinoma were higher than those in HSIL and LSIL groups(P<0.05).LSIL and HSIL were higher than those in chronic cervicitis group(P<0.05);The data were assessed by the Spearman correlation coefficients,?=0.840,bilateral test P<0.05.Showed that the level of Ki-67 protein and the range of staining increased gradually with the increase of cervical lesions.5 The expression of SCCA in cervical tissue was not correlated with HR-HPV infection(P>0.05).6 The expression of SCCA,MTA1,P16 and Ki-67 was positively correlated with HR-HPV infection(P<0.05).Conclusions 1 The expression of SCCA in cervical squamous cell carcinoma was higher than that in HSIL and LSIL group,HSIL and LSIL group were higher than that in chronic cervicitis group.2 The expression of MTA1 in cervical squamous cell carcinoma was higher than that in HSIL group,HSIL group was higher than that in LSIL and chronic cervicitis.3 P16,Ki-67 protein in cervical squamous cell carcinoma was significantly higher than that in HSIL and LSIL group,HSIL and LSIL group were higher than that in chronic cervicitis group and normal cervical group.4 The expression of SCCA in cervical tissue was not correlated with HR-HPV infection.5 The expression of MTA1,P16 and Ki-67 in cervical tissues was positively correlated with HR-HPV infection.
