Objective:To study the effect of Simo decoction(SD) on neurotensin(NT), calaitonin gene related peptide(CGPR) in serum and the receptor expression in stomach intestine,brain tissue of mouse with gastrointestinal functional disturbance by chronic stress, and to explore the mechanism of action about SD to FGIDs.Methods:The unpredictable chronic stress model was established by exerting the factors of irregular diet,reverse of day and night,binding and stimulating tails.To observe the general appearance,water intake,dietary allowance,body mass,the function of gastric emptying and small intestinal propulsion, the contents of MTL,Gas in blood serum,and pathological examination of gastrointestinal tissue of mouse for evaluation model. And then random treated with distilled water,domperidone and SD after the model succeed. The contents change of NT and CGRP in serum was tested by ELISA.The expressions of NT,CGRP in stomach intestine and brain tissue were measured by immunohistochemical staining. Result:1. With the unpredictable chronic stress, the mouse weight significantly decreased.The function of gastrointestinal motility and the contents of MTL,Gas in serum are to lower,but gastrointestinal tissue has not obviously pathology change.2. The SD can reduce the contents of NT and CGRP in serum of mouse with chronic stress,and the difference is significant compared with model group(P<0.05 or P<0.01),and has not significant compared with domperidone group(P>0.05).3. The SD can cut down the positive fiber expression of NT and CGRP in sinuses ventriculi of mouse with chronic stress.The average optical density of immune positive result is below model group(P<0.05),and compared with domperidone group there was not significant difference(P >0.05).4. The SD can reduce the positive fiber expression of NT and CGRP in duodenum tissue of mouse with chronic stress. The average optical density of immune positive result has statistics differences compared with the model group(P<0.05),and has not statistics differences compared with domperidone group(P>0.05). 5. The SD can cut down the expression of NT and CGRP in brain tissue of mouse with chronic stress. The positive cell population is less than model group(P<0.05 or P<0.05),and has not significant compared with domperidone group(P>0.05).Conclusion:1. The chronic stress can establish the model of mouse with gastrointestinal functional disturbance.2. The SD can promote gastrointestinal power.3. The SD can make the inordinate contents of NT and CGRP in serum return to normal,which indicate that it therapy FGIDs through regulating the level changes of brain-gut petide.4. The SD can regulate the receptor expression of NT and CGRP in stomach intestine,brain tissue, which is one of possible mechanism to therapy FGIDs.
