Objective:Observed Simotang active ingredient (arecoline, obtusiloba ether lactone,hesperidin and Costunolide) on gastrointestinal motility in mice with chronic stress function and serum Ghrelin, NO, Leptin levels and brain tissue Ghrelin, NO, Leptin expression. Explore the best combination of efficacy of the active ingredient, and possible mechanisms.Methods:144Kunming mice were randomly divided into normal group (8) and model group (136), the model mice were given chronic unpredictable stress modeling21d after randomly divided into model group, group Simotang and Simotang component group (15groups) of17group; modeling the first15days after starting administration7d, detect sugar consumption for each group of mice; perform gastric emptying and intestinal propulsion experiments; ELISA assay of serum NO, Ghrelin, Leptin levels; immunohistochemistry assay of brain tissue Ghrelin, NO, Leptin levelsResults:1. model group decreased sugar consumption in mice with the normal group and Simotang group differences were significant statistically significant (P<0.01); Simotang three active ingredients of the drug and four-drug group (11-15group) increase in sugar consumption in mice, and Simotang groups showed no significant difference (P>0.05), compared with model group differences were statistically significant (P <0.01); while Simotang active ingredient monotherapy and two drug combination group (1-10group) sugar consumption in mice did not significantly improve the Simotang group still has a statistically significant difference (P<0.01), with the model group, the difference was not statistically significant (P>0.05).2. model group increased gastric residual rate, intestinal propulsive rate decreased, and the normal group and Simotang group differences were significant statistically significant (P<0.01). Simotang three active ingredients of drugs and four drug combinations to reduce gastric residual rate, higher rates of intestinal propulsive, Simotang groups with no significant difference (P>0.05), compared with model group differences were statistically significance (P<0.05or P<0.01); while the residual rate of stomach and intestinal propulsion rate Simotang active ingredient monotherapy group and pairwise combinations with Simotang group still has a statistically significant difference (P<0.01), with the model group, the difference was not statistically significant (P>0.05).3. Reduce levels of serum Ghrelin levels, elevated NO, Leptin levels, with the normal group and Simotang group differences were significant statistically significant (P<0.01). Elevated Simotang active ingredient combination of three drugs and four drug groups Ghrelin levels, reduce NO, Leptin levels, and Simotang no statistically significant difference between groups (P>0.05), compared with model group differences were statistically significance (P<0.05or P<0.01); while Simotang monotherapy group active ingredients and combinations of two groups Ghrelin levels slightly elevated, NO, Leptin levels decreased slightly, with Simotang group differences were statistically significant (P<0.05or P<0.01), with the model group, the difference was not statistically significant (P>0.05).4. lower Ghrelin levels in brain tissue model, elevated NO, Leptin levels, with the normal group and Simotang were significant differences statistically significant (P<0.01); Simotang three active ingredients of drugs and IV drug combination group (table711-15group) Ghrelin levels rise, reducing nO, Leptin levels, and Simotang groups showed no significant difference (P>0.05), compared with model group differences were statistically significance (P<0.05or P<0.01).Conclusion:(1)Description the incidence of FD related with Ghrelin levels reduce, NO, Leptin levels elevated.(2) Arecoline, obtusiloba ether lactone, hesperidin, Costunolide may Simotang effective material basis, can effectively increase the chronic unpredictable stress in mice sugar consumption can be improved to varying degrees gastrointestinal motor function in mice with chronic stress.(3) Simotang these four active ingredients hesperidin, obtusiloba ether lactone, arecoline,Costunolide FD treatment mechanism of action may be related to inhibition of Leptin, NO release, promoting the release of Ghrelin related.(4)The active ingredient Simotang of its three-drug combination group and four drug combinations best effect.
