Objective:To screen out the differential expression gene from the endometrium during implantation window in patients with PCOS.And identify the marker genes in order to guide the clinic work.Methodes:The patiens with PCOS (n=6)and infertility women due to obstruction of fallopian tube(n=6) were performed endometrial biopsies during implantation window.The gene of differential expression were screened with microarrays Affymetrix U133A 2.0.And selected FADD which was down-regulated and Bcl-2 which was up-regulated to identify the result by Immunohistochemical staining and TUNEL.And to investigate the relationship with the serum E2/P levels.Results:There are differences in gene espression which the ration values higher than 2 or lower than -2.The genes which expressed were higher than 2 were the increased gene;while the genes which expressed were lower than -2 were the declined gene.Ratio values between 2 and -2, expressed almost no difference.There were one hundred and ninety-four genes of differential expresion were screened,among which 102 were up-redulated gene and 92 were down-regulated gene.There were nine genes associated with apoptosis,among which 5 were up-regelated and 4 were down-regulated,included FADD and Bcl-2.The MOD of Bcl-2 in endometrium during implantation window in patients with PCOS(0.203±0.225) was higher than normal group(0.035±0.009)by Immunohistochemical staining.Bcl-2 was correlated with the serum E2 levels. The MOD of FADD in endometrium during implantation window in patients with PCOS(0.057±0.012) was lower than normal group(0.105±0.033)by Immunohistochemical staining.FADD was correlated with the serum E2 levels.The ratio of apoptosis's cells of PCOS(0.17±0.08) group by TUNEL was lower the the normal group(0.39±0.07).Conclusion:The gene expresssion profiles of the endometrium during implantation window was affected by PCOS.And the lower ratio of apoptosis's cells may be the cause of suboptimal endometrial receptivity.The endometrial receptivity may be improved by selecting a proprated plan in clinic work.
