The FADD Gene Silencing Apoptotic Mechanisms Of Parkinson's Disease

Objective:To explore the affection of Fas-associated death domain(FADD) SiRNA on FADD,tyrosine hydroxylase(TH) on Parkinson's disease modal.Method:A total of 40 male, Wistar rats were randomly assigned to four group, with 10 rats in each group. PD group:ten rats were prepared to be hemiparkinsonian models induced by unilateral stereotaxic infusion of 6-hydroxydopamine(6-OHDA) in right substantia nigra of midbrain. FADD SiRNA group:FADD SiRNA sequences were separately injected into the right substantia nigra of SiRNA, then infusion of 6-OHDA. SiRNA negative control groups:SiRNA negative control sequences were separately injected into the right substantia nigra of SiRNA, and then infusion of 6-OHDA; The control group:rats were treated with a mixture of ascorbic acid and normal saline. Four weeks after operation, Finally, we compared the expression of FADD and TH mRNA in the right substantia nigra through in situ hybridization,compared the protein expression of FADD and TH in the right substantia nigra through immunohistochemical.Results:Compared to normal group, the protein expression of FADD,the expression of FADD mRNA in PD significantly increase; compared to PD group, the protein expression of FADD,the expression of FADD mRNA in FADD SiRNA group significantly decrease. Compared to normal group, the protein expression of TH,the expression of TH mRNA in PD significantly decrease; compared to PD group, the protein expression of TH,the expression of TH mRNA in FADD SiRNA group significantly increase.Conclusion:FADD SiRNA may inhibit DA neuron apoptosis to improve TH express, so FADD SiRNA may have protection and therapy role.
