Differential Expression Of Rac-1 In Endometrium And Clinical Observations Of Patients With Pcos Combined With IR After The Treatment On Tanshinone

Objection:Investigating the differential expression of Rac-1 in the endometrial tissue of patients with PCOS combined with IR and their level of serum testosterone(T)and LH/FSH after the treatment on tanshinone,it is to explore the related gene target of tanshinone in the treatment of PCOS and it’s role of serum T value and LH/FSH,at the same time to give new ideas and theoretical basis of Chinese medicine for the treatment of PCOS with IR with endometrial lesions in patients.Methods The patients were treated with PCOS combined with IR in the Huai’an Maternal and Child Health Hospital of China.The endometrial tissue and serum were collected from the patients’ system(tanshinone capsule)before and after 3 months treatment.The levels of Rac-1 in the endometrium were detected by qRT-PCR.The levels of T and LH/FSH were measured by ELISA and electrochemiluminescence.The experimental data were analyzed by t test method for data processing and statistical analysis,according to P<0.05 to determine the statistical significance,Results Serum steroid hormone levels were significantly higher in patients with PCOS combined with IR after tanshinone treatment(P<0.05).Results(1)Expression of endometrial tissue:The expression of Rac-1 was decreased after treatment with tanshinone in PCOS patients with IR(P<0.05),(2)Changes of serum steroid hormone levels:Serum T value and LH/FSH were higher in patients with PCOS combined with tanshinone treatment,and LH and T values were significantly lower(P<0.05),and LH/FSH goes to normal(≤ 2.5)level(P<0.05).Conclusion(1)Tanshinone in the treatment of PCOS with IR patients has a downward regulatory effect on the endometrial disease related gene Rac-1,clearing the tanshinone in the clinical treatment of the target.(2)Tanshinone has the effect of lowering serum T value and LH level,which can significantly improve the regulation of LH/FSH,and can effectively improve the symptoms of insulin resistance and endometrial lesions in PCOS patients with IR.
