Objective:To investigate the effect of Achyranthes bidentata on the expression of nest protein (Nestin) and growth associated protein-43 (GAP-43) in the brain of rats with severe brain injury, and explore the effect of Achyranthes bidentata on central nerve regeneration of rats with severe brain injury.Methods:According to self-made improvement Feeney method, prepare severe brain injury model, then the 65 SD rats were randomly divided into Achyranthes bidentata high dose group,Achyranthes bidentata middle dose group,Achyranthes bidentata low dose group,model group and sham operation group, Each group of 13 only. The rats were sacrificed for the brains after 7 days of treatment, then the brains were normally made into paraffin. brains tissue's pathological changes were studied by HE staining, The expressions of Nestin and GAP-43 were determined by immunohistochemistry staining.Results:1.The scores of Nestin in high dose group were increased significantly,compared with the model group, the difference is significant (P<0.05); there was not significant difference in Nestin between middle dose group, low dose group and model group; very little expression of Nestin-positive cells were occasionally found in hippocampus of sham operation group, some sections didn't have positive expression; 2. The scores of GAP-43 in high dose group were significantly higher than the model group (P<0.05); there was not significant difference in GAP-43 between middle dose group, low dose group and model group; the expression of GAP-43 in sham operation group was lower than the other groups(P<0.05).Conclusion:High dose Achyranthes bidentata can raise the expression of Nestin and GAP-43 in rat brains with severe brain injury, possibly promote the regeneration of Central nervous through increased expression of Nestin and GAP-43 after severe brain injury.
