Objective Severe brain injury models were established in adult rats by free falling objects.To study effects of differrent doses of Achyranthes Bidentata Blume on the interleukin-2 (IL-2) of blood serum, the soluble interleukin-2 receptor (sIL-2R) of blood serum, the phagocytic function of PMN in rats with severe brain injury, and explore Achyranthes Bidentata Blume on immune regulation in rats with severe brain injury.Methods SD rats with body weight of 220g±30g were randomly divided into six groups (n=13).Seventy-eight rats were averagely matched as the sham injury group, the model group, the high-dose group, middle dose group, low-dose group of Achyranthes Bidentata Blume and the Naloxone group.1. Severe brain injury model were established in adult rats by free falling objects.2. The neural function of rats injuried were scored after 6 hours3. Specimens of serum and anticoagulant heparin and brain tissue were collected respectively on the 7th day after injury.4. The level of IL-2 and sIL-2R in SD rats was analyzed by enzyme-linked immunosorbent assay (Elisa testing).5. The PMN phagocytic function in peripheral blood was detected by the technique of immunology and cytology.6. Normally paraffin sections of brain tissue were stained with HE7.The areas of lesion and surrounding tissue were observed by microscope,and image analysis software was used to total the area of damage.8. Laboratory animals used in this study have been reviewed by the Ethics Committee of Guangxi Medical University.Results1. The score of neural function of these rats injured and general moephology of the brain and the brain tissue pathology with microscopic observation showed that the models of severe brain injury of rats were made successfully.2. Serum level of IL-2 was obviously decreased in the model group, low-dose group and naloxone group, which were respectively compared with the sham injury group and high-dose group, and the differences were significant (P<0.01).Compared with the model group and low-dose group respectively,there were significant differences in the middle dose group (P<0.05). There were no significant differences (P>0.05)among other groups.3. Serum level of sIL-2R was obviously higher in the model group than in the sham injury group and high-dose group, and the differences were significant (P<0.05). There were no significant differences (P>0.05) among other groups.4.Phagocytic rate of PMN was obviously decreased in the model group, middle dose group and low-dose group which compared with sham-injury group, and the differences were significant (P<0.01). High-dose group respectively compared with the model group and low-dose group, and the differences were significant (P<0.05). Other groups compared with each other, and there were no significant differences (P>0.05).Conclusion1. Severe brain injury model was established successfully in adult rats by self-made modified free falling objects. Its operation was simple and strength of impact could be controlled easily. The size of the injury area was stable and the repetition was good and the survival rate of rats was high.2. Immune function of rats were depressed after severe brain injury.Middle-dose and high-dose decoction of Achyranthes Bidentata Blume could improve inhibition of immune function,especially in high-dose decoction of Achyranthes Bidentata Blume.3. Achyranthes Bidentata Blume on immune regulation in rats with severe brain injury,on the one hand itself could enhance the immune function,on the other hand could also improve the central nervous system disorders related to immune regulation.
