Objective: To observe the treatment effect of moisture content on brainorganization and AQP4expression of severe brain injury rats by usingtraditional Chinese medicine achyranthes bidentata and astragalusmembranaceus;and to research the mechanism of curing severe brain edema。Methods: Sixty five SD rats are divided randomly into sham-operation group(5),model group(15),achyranthes bidentata treated group(15),astragalusmembranaceus treated group (15), achyranthes bidentata and astragalusmembranaceus treated group(15).Then to set the severe brain injury model inrats by using improved Feency’s methods. After that, to measure the moisturecontent on brain organization, to observe the changeable condition by using HEstaining,and to detect the brain organization on AQP4expression by usingimmunohistochemical ways on the first day, the third day and the seventh day.Result: On each timing measure, the moisture content on brain organizationand the AQP4expression around the injury part of the model group rats arehigher than the sham-operation group(P<0.05), and the brain edema is obviously changed on the model group after using HE. staining While theachyranthes bidentata treated group and the astragalus membranaceustreated group have no obvious reduction (P>0.05), and HE staining are the sameas the model group. Then in the achyranthes bidentata and astragalusmembranaceus treated group, the moisture content and the AQP4expression arelower than the model group(P<0.05), and compared to the model group, thebrain edema has a improved change after using HE staining.Conclusion:1.Severe brain injure model was established in adult rats by self-mademodified free falling objects. the method is simple, reliable, reproducible andlow mortabity.2. The development of severe brain injury brain edema has the relation withthe AQP4of brain tissue.3.The brain edema caused by severe brain injury can be improved in theachyranthes bidentata and astragalus membranaceus treated group. Its effectmay has a relation with the expression of AQP4restraint in injury brainorganization and the reduction of injury on brain cells.
