Objective:The subject through the establishment of mouse models of exhaustive swimming to prove the teacher's hypothesis of the "fatigue" factor of "Joint effects of body shock" theory in the body caused by the "cell organic change" and to observate in heart, liver, spleen, lung, kidney, brain, stomach, intestine, thymus, pancreas, adrenal gland, skeletal muscle, testis tissue pathological changes after the exhaustive swimming and the 4th day after termination of the fatigue factor under light microscope and electron microscope changes in mitochondriaMethods:By random number table the mice were divided into three groups:10 of the control group; 10 of the first group of exhausted swimming; 10 of the second group of the exhausted swimming.Traditional Chinese Medicine in adaptive breeding animals for three days and were loaded on the third day for the exhaustive swimming test:To weigh the weight of each mouse and each mouse was loaded for 5% of their body weight.And put them in glasscylinder fill water with the depth of 50cm and the temperature (25±1)℃and swim to exhaustion.When 10s can not be submerged in the water surface that is judged to be exhausted state.At the end of exhaustive swimming to kill the mice of the control group and the first group of exhausted swimming and the mice of the second group of the exhausted swimming were resumed three days and killed on the fourth day.Remove the heart, liver, spleen, lung, kidney, brain, stomach, intestinal, thymus, pancreas, adrenal gland, skeletal muscle and testis of the mouse and was cut in two parts, respectively, which in the 10% formaldehyde solution and the other in the 4% glutaraldehyde solution.The tissue in the 10% Formaldehyde solution were washed, graded ethanol dehydration, paraffin embedding soaked, sliced, stained by HE,sticked by neutral gum and watched in light microscopy.The tissue in the 4% glutaraldehyde solution were washed by the PBS,fixed by osmium tetroxide,washed by double distilled water, gradiented dehydration by acetone, saturated embedding by 812 resin,sliced in advanced micro devices, stained and watched in electron microscopy.Results:1.The results of experimental show that the state of exhaustive swimming on the body of mice were burnout and can be recovery in 5 minutes and self-grooming and returned to normal state in 30 minutes in the general.2.The exhaustive swimming test in mice which caused by fatigue factors on the body appeared burnout phenomenon and the joint effect of the body caused by the test cause multi-system shock and multiple organ tissue pathological changes of different degrees of atrophy, edema and fatty change.The test also leading to multi-system and multi-organ tissue cells showed the organic changes of the number of mitochondria reduced and mitochondria shorter or disappeared and the ridge between the cavity expansion.3.The test shows when the fatigue factors were eliminated and the burnout will resume in a short period of time and improve clinical symptoms.But when the fatigue factors were eliminated and most of the cells were not only failed to reduce the organic change but heavier after 4 days.4.The mitochondria, especially mitochondrial 8m of some of the few animal organ tissue in the control group of the exhaustive swimming test also appeared the organic changes of shorter or disappeared and the ridge between the cavity expansion.Conclusion:1.The result confirmed the hypothesis which Proposed by the tumor the joint effect of the body caused by some of the factors cause the organic changes of Multiple organs in the body.2.This phenomenon may explain the recovery of clinical symptoms caused in a short time can be significantly improved or basically return to normal, but the organic change of cells can not be quickly restored.3.The fatigue factors were eliminated and most of the cells were not only failed to reduce the organic change but heavier after 4 days. may be Related with oxygen free radicals increased, SOD decreased, calcium overload, ATP decreased and remains to be further studied...
